Rubus rosaefolius Extract as a Natural Preservative Candidate in Topical Formulations

Even though the synthetic preservatives may offer a high antimicrobial efficacy, they are commonly related to adverse reactions and regarded as having potentially harmful effects caused by chronic consumption. The development of natural preservatives provides a way of reducing the amount of synthetic preservatives normally used in pharmaceutical and cosmetic preparations. In addition, these agents have less toxic effects and represent a possible natural and safer alternative of the preservatives. The purpose of this research was to evaluate the Rubus rosaefolius Smith extract efficiency as a natural preservative in base formulations. Of the extract, 0.2% (w/w) was assayed for its effectiveness of antimicrobial protection in two different base formulations (emulsion and gel). The microbial challenge test was performed following the standard procedures proposed by The United States Pharmacopoeia 33nd, European Pharmacopoeia 6th, Japanese Pharmacopoeia 15th, and the Cosmetics, Toiletries, and Fragrance Association using standardized microorganisms. The emergence of less common fungal pathogens has been increasingly reported in the last decade. We describe 25 cases of Rhodotorula spp. isolated from blood cultures at a large Brazilian tertiary teaching hospital from 1996-2004. We also investigated the in vitro activity of four antifungal drugs, using a standardized method. The median age of patients was 43 years. The majority of patients (88%) had a central venous catheter (CVC) and 10 (40%) were recipients of a bone marrow transplant. The episode was classified as a bloodstream infection (BSI) in 80% of the patients. Amphotericin B deoxycholate was the most common antifungal used and CVC was removed in 89.5% of the patients. Death occurred in four patients (17.4%), all classified as BSI. All strains were identified as R. mucilaginosa by conventional methods. Misidentification of the species was observed in 20% and 5% of the strains with the Vitek Yeast Biochemical Card and API 20C AUX systems, respectively. Amphotericin B demonstrated good in vitro activity (MIC(50/90), 0.5 mu g/ml) and the MICs for fluconazole were high for all strains (MIC(50/90), 64 mu g/ml).

A técnica de análises PIXE (Particle Induced X-ray Emission) de alvos finos é rotineiramente usado no Instituto de Física da Universidade de São Paulo (USP) pelo Lamfi (Laboratório de Materiais e Feixes Iônicos) em análises quantitativas elementares. A calibração do arranjo experimental do Lamfi é realizada através da irradiação de filmes finos padrões evaporados, sendo o rendimento de produção de raios X ajustado a partir deprimeiros princípios. Em análises PIXE de alvos espessos (TTPIXE), a composição daparticular amostra introduz efeitos de perda de energia das partículas incidentes e autoabsorção dos raios X produzidos. Estes efeitos são inexistentes na análise PIXE de alvosfinos. Paradoxalmente há a necessidade de se conhecer a composição da amostra, paracomputar estes efeitos, numa análise elementar quantitativa de alvos espessos. Nos últimos anos a eletroforese capilar tem substituído a eletroforese em gel e está sendo usada para uma ampla variedade de aplicações forenses, incluindo tipagem de DNA. A fim de superar as desvantagens com relação à análise simultânea de amostras que a eletroforese em gel oferece, equipamentos com arranjos de capilares foram idealizados, assim como a possibilidade de análises multiplexadas em um único capilar por meio da utilização de corantes intercaladores. Neste trabalho foi otimizada a metodologia para amplificação de DNA pela reação em cadeia da polimerase para sete primers correspondentes a sete regiões padronizadas e legalmente aceitas para testes de paternidade. Três casos foram avaliados por eletroforese em microchip, indicando que um método mais reprodutível e de maior resolução deveria ser utilizado, fato que levou ao desenvolvimento de um método para separação de um padrão de tamanho de DNA de 25 pares de base por eletroforese capilar em soluções poliméricas em um equipamento comercial. Existem evidências na literatura de que a administração de medicações, como corticosteróides inalatórios e broncodilatadores, utilizando inaladores pressurizados dosimetrados, é mais eficaz e segura do que com os nebulizadores convencionais. Entretanto, há evidências de que não só pacientes, mas também profissionais de saúde têm dificuldade no manejo dos inaladores pressurizados dosimetrados. A hipótese do presente estudo é que o conhecimento da técnica inalatória entre os profissionais da nossa Instituição é heterogêneo, sendo que não há informações sobre o nível deste conhecimento entre as diversas categorias profissionais. OBJETIVOS: Avaliar o conhecimento prático e teórico de profissionais da área médica, de enfermagem e fisioterapia sobre a utilização do inalador dosimetrado através de testes padronizados. O objetivo desta dissertação é escrever sobre a técnica de ultra-sonografia, aplicada na inspeção da soldagem por resistência a ponto, e demonstrar as vantagens desta técnica em relação aos ensaios destrutivos convencionalmente normalizados e utilizados na inspeção de solda a ponto até meados de 1999, quando a técnica de ultra-som começou a ser aplicada no Brasil. Para tanto foram realizados dois procedimentos experimentais, sendo o primeiro deles utilizando a técnica de ultra-som a-scan (transdutor de simples elemento), onde foram estudados os efeitos da indentação do ponto de solda, um dos parâmetros que pode ser detectado com extrema facilidade por esta técnica. O segundo experimento visa demonstrar a capacidade de detecção da técnica de ultra-som b-scan (transdutor matricial), onde a metodologia foi aplicada visando correlacionar os resultados obtidos pelo equipamento com os resultados encontrados nas técnicas de arrancamento (peel test) e nos ensaios macrográficos convencionalmente conhecidos. Although delayed-type hypersensitivity skin testing with tuberculin purified protein derivative (PPD) is the standard for tuberculosis screening, its variability suggests the need for a more sensitive, noninvasive test. An in vitro whole-blood assay has been proposed as an alternative. Using health care worker volunteers, we confirmed the correlation between PPD skin test (PPD-ST) results (positive, induration of >15 mm) and a standardized gamma interferon (IFN-γ) assay, QuantiFERON-TB (Q-IFN), manufactured by CSL Biosciences in Australia, and we evaluated Mycobacterium tuberculosis culture subfractions as potential substitutes for PPD. Twenty healthy volunteers with positive PPD-ST results and 20 PPD-ST-negative controls were enrolled. Whole blood was cultured with human PPD antigens (HuPPD), Mycobacterium avium complex (MAC) PPD, phytohemagglutinin (PHA), and four M. tuberculosis culture subfractions: low-molecular-weight culture, filtrate, culture filtrate without lipoarabinomannan, soluble cell wall proteins, and cytosolic proteins, all developed from M. tuberculosis strain H37RV. Secretion of IFN-γ (expressed as international units per milliliter) was measured by an enzyme immunoassay. Objective: In the United States, around 50% of all musculoskeletal injuries are soft tissue injuries including ligaments and tendons. The objective of this study is to assess the role of amnion-derived cellular cytokine solution (ACCS) in carboxy-methyl cellulose (CMC) gel in the healing of Achilles tendon in a rat model, and to examine its effects on mechanical properties and collagen content. Methods: Achilles tendons of Sprague-Dawley rats were exposed and transected. The distal and proximal ends were injected with either saline or ACCS in CMC, in a standardized fashion, and then sutured using a Kessler technique. Tendons from both groups were collected at 1, 2, 4, 6, and 8 weeks postoperatively and assessed for material properties. Collagen studies were performed, including collagen content, collagen cross-linking, tendon hydration, and immunohistochemistry. Tendons were also evaluated histologically for cross-sectional area. Results: Mechanical testing demonstrated that treatment with ACCS in CMC significantly enhances breaking strength, ultimate tensile strength, yield strength, and Young's modulus in the tendon repair at early time points. In context, collagen content, as well as collagen cross-linking, was also significantly affected by the treatment. Conclusion: The application of ACCS in CMC has a positive effect on healing tendons by improving mechanical properties at early time points. Diagnostic material from patients with leishmaniasis is generally available as promastigotes, and proper testing for susceptibility to first-line drugs by the intracellular amastigote assay is frequently hampered by the poor infectivity of the promastigotes for the macrophage host cell. Several conditions for optimization of the in vitro metacyclogenesis and cell infectivity of Leishmania donovani, L. guyanensis, and L. braziliensis field strains obtained from patients receiving standard antimony medication were investigated. Triggering log-phase promastigotes to become amastigote-like by increasing the temperature or acidifying the culture medium was not successful. Adequate metacyclogenesis and the highest levels of macrophage infection were obtained after 5-day-old late-log-phase promastigote cultures were preconditioned at 25°C to pH 5.4 for 24 h in Schneider's medium prior to infection. The susceptibility assay with primary peritoneal mouse macrophages included pentavalent antimony (SbV; sodium stibogluconate), trivalent antimony (SbIII; potassium antimonyl tartrate), miltefosine, and the experimental drug PX-6518. Collecting the contextual factors that influence patient/client outcomes, quality, and cost of health care is essential for comparison of delivery systems and health care plans. Measuring the effects of management and administrative interventions and quantifying the characteristics of the environment within which care is delivered are essential to determining the validity of studies measuring patient/client outcomes and effectiveness. This paper describes the research based development of the Nursing Management Minimum Data Set (NMMDS) and outlines a strategy for clinical testing of the NMMDS across 50 US sites, settings, and health systems. The NMMDS, the product of a collaborative effort between The University of Iowa NMMDS Research Team and the American Organization of Nurse Executives (AONE), is grounded in nursing's history of managing, coordinating, and integrating care across sites and settings. Implications for measuring the nursing component, alone or as a part of a broader whole of health care delivery redefinition, are explained. PURPOSE: We sought to characterize the dyschromatopsia of optic neuritis, to determine the type and severity of color defect present and its relation to central vision and spatial acuity, to examine changes in this dyschromatopsia over time, and to determine the applicability of Köllner's rule to patients with optic neuritis. METHODS: We analyzed the raw data on color vision performance as assembled within the Optic Neuritis Treatment Trial (ONTT). The ONTT was designed to evaluate corticosteroids as a treatment for acute demyelinating optic neuritis and to allow long-term outcome and natural history analyses. Between July 1, 1988 and June 30, 1991, 488 patients were enrolled in this trial. All patients underwent extensive neurologic and ophthalmologic examinations including standardized testing of visual function that included testing of color vision. The ONTT population thus afforded a unique opportunity to characterize acquired dyschromatopsias in a large, homogenous, well-characterized cohort of patients with optic neuritis. We used quantitative analysis of FM-100 scores from this patient cohort to determine the severity of the dyschromatopsia, the selectivity of the dyschromatopsia (polarity of errors) and the type of dyschromatopsia (axis of confusion) by employing quadrant analysis of FM-100 scores. A study was performed in two laboratories to evaluate the effect of growth medium and test methodology on inter- and intralaboratory variations in the MICs of amphotericin B (AMB), flucytosine (5FC), fluconazole (FLU), itraconazole (ITRA), and the triazole Sch 39304 (SCH) against 14 isolates of Candida albicans. Testing was performed by broth microdilution and semisolid agar dilution with the following media, buffered to pH 7.0 with morpholinepropanesulfonic acid (MOPS): buffered yeast nitrogen base (BYNB), Eagle's minimal essential medium (EMEM), RPMI 1640 medium (RPMI), and synthetic amino acid medium for fungi (SAAMF). Inocula were standardized spectrophotometrically, and endpoints were defined by the complete absence of growth for AMB and by no more than 25% of the growth in the drug-free control for all other agents. Comparative analyses of median MICs, as determined by each test method, were made for all drug-medium combinations. Both methods yielded similar (+/- 1 twofold dilution) median MICs for AMB in EMEM and RPMI, 5FC in all media, and FLU in EMEM, RPMI, and SAAMF. A diagnosis of food allergies should be made based on the observation of allergic symptoms following the intake of suspected foods and the presence of allergen-specific IgE antibodies. The oral food challenge (OFC) test is the most reliable clinical procedure for diagnosing food allergies. Specific IgE testing of allergen components as well as classical crude allergen extracts helps to make a more specific diagnosis of food allergies. The Japanese Society of Pediatric Allergy and Clinical Immunology issued the 'Japanese Pediatric Guideline for Food Allergy 2012' to provide information regarding the standardized diagnosis and management of food allergies. This review summarizes recent progress in the diagnosis of food allergies, focusing on the use of specific IgE tests and the OFC procedure in accordance with the Japanese guidelines.

Computerized adaptive testing is becoming increasingly popular due to advancement of modern computer technology. It differs from the conventional standardized testing in that the selection of test items is tailored to individual examinee's ability level. Arising from this selection strategy is a nonlinear sequential design problem. We study, in this paper, the sequential design problem in the context of the logistic item response theory models. We show that the adaptive design obtained by maximizing the item information leads to a consistent and asymptotically normal ability estimator in the case of the Rasch model. Modifications to the maximum information approach are proposed for the two- and three-parameter logistic models. Similar asymptotic properties are established for the modified designs and the resulting estimator. In statistics, the Durbin-Watson test is always employed to detect the presence of serial correlation of residuals from a least squares regression analysis. However, the Durbin-Watson statistic is only suitable for ordered time or spatial series. If the variables comprise cross-sectional data coming from spatial random sampling, the Durbin-Watson will be ineffectual because the value of Durbin-Watson's statistic depends on the sequences of data point arrangement. Based on the ideas from spatial autocorrelation, this paper presents two new statistics for testing serial correlation of residuals from least squares regression based on spatial samples. By analogy with the new form of Moran's index, an autocorrelation coefficient is defined with a standardized residual vector and a normalized spatial weight matrix. Then on the analogy of the Durbin-Watson statistic, a serial correlation index is constructed. As a case, the two statistics are applied to the spatial sample of 29 China's regions. These results show that the new spatial autocorrelation model can be used to test the serial correlation of residuals from regression analysis. In an effort to construct the most advanced school system in the nation, Los Angeles school administrators and educators initiated a new scientific method of group intelligence testing. Almost immediately educators discovered serious limitations with the process and resisted its exclusive use. This study examines the reception of this new technology in Los Angeles between 1922 and 1932. Many historians have seen those associated with I.Q. measuring as bulwarks supporting the hegemony of Anglo-Saxon upper-middle class society. While their criticism has brought some non-equitable aspects of twentieth-century public education to surface, it has not led to our understanding of how educators interpreted the tests. An analysis of the sources, including reports published in the Department of Psychology and Education Research Bulletin of the Los Angeles City Schools, the Teachers' and Principals' School Journal, and the Minute~ of the Board of Education, provides insight into how Los Angeles educators viewed standardized testing.

Este artigo discute a importância do pré-teste e os resultados da sua aplicação num procedimento para avaliação de desempenho de empreendimentos habitacionais em uso, visando contribuir para o conhecimento sobre a qualidade do ambiente construído e dos sistemas construtivos inovadores especificados dentro de uma visão sistêmica do ciclo de vida útil das edificações.  O procedimento padronizado englobando um conjunto de instrumentos otimizados para avaliação de desempenho, ora proposto, teve como base a metodologia da Avaliação Pós-Ocupação (APO) e os requisitos e critérios da norma brasileira ABNT/NBR 15575:2013.  Ressalta-se que esse conjunto de instrumentos foi concebido para permitir uma avaliação preliminar do atendimento do sistema construtivo inovador de empreendimentos habitacionais aos requisitos de desempenho definidos em normas brasileiras, considerando as suas principais características.  Os instrumentos propostos e pré-testados indicam ser fundamental a participação dos usuários (moradores) na avaliação das habitações em fase de uso, combinada com a aplicação de instrumentos para a avaliação técnica de desempenho realizada por especialistas, como forma de obtenção de insumos essenciais para aprimorar o processo e a gestão visando à garantia da qualidade arquitetônica e construtiva.; This article discusses the importance of pre-testing and the results of itsapplication in a procedure for performance evaluation of occupied residential buildings tocontribute for improving the knowledge about the quality of both the built environment andthe innovative building systems specified within a systemic view of the life cycle of buildings.The proposed standardized procedure...

This dissertation consists of three separate essays on the economics of education. In the first chapter, co-authored with Esteban Aucejo, studies the relative effectiveness of reducing absences to extending the school calendar on test score performance. Using administrative data for North Carolina public schools, we exploit a state policy that provides variation in the number of days prior to standardized testing and find substantially larger effects for absences relative to additional days of class.

The second chapter, co-authored with Esteban Aucejo, analyzes whether different institutional settings could affect how school administrators and teachers respond to possible extensions of the school calendar. We present a theoretical model in which principals set the date of the test and teachers decide how much effort to exert in the classroom with and without monetary performance bonuses for teachers. Leveraging the removal of monetary bonuses during the sample period, we utilize a difference-in- difference estimation strategy and find that, consistent with the theoretical model, low performing schools are more likely to make extensive use of the testing window when monetary bonuses are in place; this behavior disappears after changes to the scheme of incentives.

Business schools: still in the dark during the ‘Age of enlightenment’

With the establishment of emotional intelligence (EI) and EI testing, psychology has offered the business world a powerful and invaluable tool capable of predicting success in the workplace. The most progressive organizations are already employing such tests in their HR selection/promotion processes. The business school industry maintains a vital interface with the various industry sectors, providing companies with their ‘senior executives of the future’. However, even given the highly competitive environment in which business schools operate; attracting top MBA candidates, and later placing their graduates with prestigious companies, they continue utilizing an outdated, IQ-based, standardized test (the GMAT) as a critical, and often times deciding criterion within their graduate selection processes. The recognized limitations of IQ, as well as its failure to predict professional success, remain a catalyst for the popularization of EI. The author proposes that, in conjunction with top recruiting organizations, business schools initiate an EI-based ‘soft skills’ admission test that would compliment, or even subsume the existing Graduate Management Admission Test.
