Antigenicity of a whole parasite vaccine as promising candidate against canine leishmaniasis

Human visceral leishmaniasis, one of the most important zoonoses, is caused by the protozoaLeishmania chagasi(syn. L. infantum ) and is present as a fatal disease common in South America and Europe where dogs and wild canids are the main reservoirs. A vaccine against visceral leishmaniasis would be an important tool in the control of this disease in dogs. Although the current strategies for vac-cination against leishmaniasis are based on the use of recombinant antigens, killed vaccines are still attractive in terms of stability of their biochemical composition and antigenicity, cost, and safety. Here we evaluate the immunogenicity of a whole parasite vaccine as a prom-ising candidate against canine leishmaniasis, demonstrated by cellular reactivity, changes in the cellular profile of the peripheral blood and by the differential production of immunoglobulins. Our results showed that immunization elicited mainly a strong cellular reactivity and increase in T-lymphocytes, particularly the subpopulation CD8 + that would be related to the control of tissue parasitism. In addi-tion, a higher production of anti- Leishmania total IgG, characterized by mixed isotypes profile (IgG1 and IgG2), was demonstrated.

The kinetics and microstructural evolution of austenite formation in a low carbon steel, with initial microstructure composed of ferrite and pearlite, were studied during continuous heating, by using dilatometric analysis and measurements of microstructural parameters. The formation of austenite was observed to occur in two stages: (a) pearlite dissolution and (b) ferrite to austenite transformation. The critical temperatures of austenite formation in continuous heating increase with increasing heating rate, with greater influence on the finishing temperature of austenite formation. For both the 1 ?C/s and 0.1 ?C/s heating rates, the formation rate of austenite reaches a maximum at approximately the finishing temperature of pearlite dissolution, and the formation rate of austenite as a function of the temperature is greater at the higher heating rate.

A vaccine against canine visceral leishmaniasis (CVL), comprising Leishmania braziliensispromastigote protein, sand fly gland extract (SGE) and saponin adjuvant, was eval-uated in dog model, in order to analyse the immunogenicity of the candidate vaccine. The vaccine candidate elicited strong antigenicity in dogs in respect of specific SGE andLeishmania humoral immune response. The major saliva proteins recognized by serum from immunized dogs exhibited molecular weights of 35 and 45 kDa, and were related to the resistance pattern against Leishmaniainfection. Immunophenotypic analysis revealed increased circulating CD21 + B-cells and CD5 + T-cells, reflected by higher counts of CD4 + and CD8 + T-cells. The observed interac-tion between potential antigen-presenting cells (evaluated as CD14 + monocytes) and lymphocyte activation status indicated a relationship between innate and adaptive immune responses. The higher frequency in L. chagasi antigen-specific CD8 + T-lymphocytes, and their positive association with intense cell proliferation, in addition to the progressively higher production of serum nitric oxide levels, showed a profile compatible with anti-CVL vaccine potential. Further studies on immunological response after challenge with L. chagasi may provide important information that will lead to a better understanding on vaccine trial and efficacy.

Serological screening confirms the re-emergence of canine leishmaniosis in urban and rural areas in Governador Valadares, Vale do Rio Doce, Minas Gerais, Brazil

This study performed clinical , serological and parasitological assessments in dogs from Vale do Rio Doce , in Minas Gerais State , Brazil, a region considered as a ? controlled endemic ? area for canine visceral leishmaniosis (CVL). Nevertheless, there are signs that CVL in dogs may be re-emerging as a programme to control the disease was interrupted in the 1990s. The majority of the animals examined presented various symptoms associated with CVL. The enzyme-linked immunosorbent assay test indicated 13.7 and 12.4% of positivity of dogs from the urban and rural areas, respectively. According to indirect immunofluorescence assay test and TRA Ld test s, 18.2 and 42.2% of dogs in the rural area were seropositive, respectively. Parasitism in seropositive dogs was confirmed by in vitro tissue culture. Sand flies of the genus Lutzomyia, which are able to transmit both cutaneous and visceral leishmaniosis, were found in the area. The results provide a strong evidence of the re-emergence of CVL in this region.

O Grupo Paje? constitui a unidade superior do Supergrupo Oliveira dos Brejinhos ocorrendo na borda leste da Serra do Espinha?o Setentrional no estado da Bahia. Essa unidade corresponde ao preenchimento de uma bacia do tipo rifte, durante um dos est?gios evolutivos da Bacia Espinha?o no norte do Craton S?o Francisco. A Forma??o Riacho Fundo, unidade basal do Grupo Paje?, foi depositada predominantemente por sistemas de leques aluviais e de rios anastomosados, submetidos ao retrabalhamento e?lico. Subsequentemente, esta sucess?o foi recoberta pelos sedimentos da Forma??o Ipu?aba, depositados num sistema retrogradacional lacustrino-delt?ico. O est?gio final do preenchimento da bacia na parte sul foi caracterizado pela ocorr?ncia de vulcanismo intermedi?rio a ?cido associado ? deposi??o de sedimentos cl?sticos e vulcanocl?sticos da Forma??o Bomba, de idade eo-calamiana. The S?o Francisco Craton, in the central-eastern region of Brazil, is a cratonic fragment stabilized after the Rhyacian tectono-metamorphic event (2.25?2.05 Ga). Volcanic and sedimentary rocks of the Espinha?o (Paleo- to Mesoproterozoic) and S?o Francisco (Neoproterozoic) basins crop out on this craton. In the northern sector of the craton, the Espinhac? o basin crops out in two domains: the Northern Espinha?o Range and the Chapada Diamantina, that are separated by the Paramirim Corridor, a Brasiliano/Pan African (650?500 Ma) deformation zone, which crosses the craton in NNW?SSE direction. The stratigraphic framework of the Northern Espinhac? o is characterized by eight synthems that comprise units bound by unconformities, or stratigraphic discontinuities with regional extent over the Espinha?o basin. These synthems are: Algod?o, S?o Sim?o, Sapiranga, Paje? and Bom Retiro (lower interval), S?o Marcos and S?tio Novo (intermediate interval) and Santo Onofre (upper interval). For each interval the sedimentary processes, the depositional systems, the filling style of the basin and the tectonic settings were evaluated, based mainly on the facies association characteristics and their lateral/vertical changes. A por??o leste da Bacia do S?o Francisco, nos arredores da serra da ?gua Fria, conta com uma se??o s?smica (240-RL-300) de dire??o NW-SE. Ao longo dessa se??o foi poss?vel reconhecer os estilos estruturais dominantes no segmento estudado e constatar que o mesmo pertence a um compartimento tect?nico, denominado Sali?ncia de Pirapora, onde o embasamento foi envolvido na deforma??o das rochas da cobertura (tect?nica thickskinned). A partir da integra??o dos dados s?smicos e de campo, foi poss?vel dividir esse segmento em 3 dom?nios estruturais: Oriental, Central e Ocidental, que s?o caracterizados, no n?vel estrutural mais raso, pela presen?a de megadobras, uma extensa monoclinal (Monoclinal do Boqueir?o) e estratos suborizontais, respectivamente. A deforma??o ? maior no dom?nio Oriental e diminui em dire??o ao Ocidental. Objectives: To evaluate the effects of benznidazole on Chagas? disease cardiac prognosis using an experimental dog model of infection. Methods: A total of 28 dogs were divided into three groups: 10 were infected with Trypanosoma cruzi and treated benznidazole during the chronic phase, 10 were infected but untreated, and 8 were non-infected/ healthy. The trypanocidal efficacy was measured by parasite kDNA detection in blood and cardiac tissue samples. The effects of benznidazole in ameliorating the cardiac systolic function were evaluated by echodopplercardiogram. Results: The benznidazole initially induced a potent suppression of parasitaemia in treated animals. However, 12 months post-treatment, the parasite kDNA detections were similar between infected groups. In the baseline echocardiographic parameters there was no variation among all animals. Similarly, 1 month post-treatment there was no significant difference among healthy and infected animals with regard to systolic function. At 12 months post-treatment, an increase in cardiac chamber size related to cardiomegaly was detected among treated and untreated animals, but not in the healthy controls. OBJETIVO: Analisar os aspectos ?ticos dos artigos nacionais sobre leishmaniose visceral humana publicados ap?s a Resolu??o CNS 196/1996 e analisar a pol?tica de peri?dicos brasileiros sobre ?tica em pesquisa. M?TODOS: Estudo explorat?rio de natureza bibliogr?fi ca e documental. A sele??o das publica??es sobre pesquisas envolvendo seres humanos foi realizada na base SciELO Brasil a partir de 1996. Foram analisadas lacunas relacionadas ?s pol?ticas editoriais dos peri?dicos m?dicos a partir de informa??es obtidas da se??o ?Instru??es aos autores? de cada peri?dico. RESULTADOS: Observou-se que, enquanto no primeiro quadri?nio (1997 a 2000) nenhum artigo mencionou o cumprimento dos aspectos ?ticos, no ano de 2009 do quadri?nio (2009 a 2012), 75% cumpriram pelo menos uma das orienta??es ?ticas avaliadas. Seis em 11 peri?dicos indicaram que as informa??es sobre os aspectos ?ticos deveriam ser citadas no corpo do artigo. Tr?s peri?dicos fi zeram exig?ncias quanto ao envio de carta ou documento assinado pelo(s) autor(es) informando o cumprimento dos aspectos; dois solicitaram c?pia do documento de obten??o do termo de consentimento livre e esclarecido; um explicitou a necessidade do envio de c?pia da autoriza??o da aprova??o pelo comit? de ?tica em pesquisa; e em quatro peri?dicos analisados n?o foi identifi cada nenhuma exig?ncia sobre aspectos ?ticos. CONCLUS?ES: Houve melhora em rela??o ? descri??o do cumprimento dos aspectos ?ticos nos artigos. Sugere-se a padroniza??o das orienta??es ?ticas relacionadas ? pesquisa com seres humanos nos peri?dicos nacionais. In this work, we investigated the in vivo activity of ravuconazole against the Y and Berenice-78 Trypanosoma cruzi strains using acutely infected dogs as hosts. Ravuconazole was well tolerated, as no significant side effects were observed during the treatment using 6.0 mg/kg twice a day (12 mg/kg/day) for up to 90 days. In all treated animals, parasitemia was permanently suppressed by the first day of treatment, independently of the parasite strain. Cultures of blood obtained posttreatment were negative for 90% of the animals, confirming that the drug induced a marked reduction in the parasite load. The results of PCR tests for T. cruzi in blood performed 1 month posttreatment were consistently negative for three of five and two of five animals infected with the Y and Berenice-78 strains, respectively. All ravuconazole-treated dogs consistently had negative serological test results during and until 30 days after treatment, regardless of the therapeutic scheme used. Asthma and chronic obstructive pulmonary disease (COPD) are common respiratory illnesses characterized by chronic inflammation of the airways. The characterization of induced or spontaneously produced sputum is a useful technique to assess airway inflammation. In the present study, we compared the concentrations of CCL2, CCL11, CXCL8, and tumor necrosis factor-??(TNF-?) in plasma and induced sputum of patients with severe asthma or COPD and correlated the levels of these mediators with inflammatory cells in sputum. Asthmatic patients had elevated levels of eosinophils (40.1 ? 6.24%) in sputum whereas neutrophils (63.3 ? 4.66%) predominated in COPD patients. The levels of the chemokine CCL11 were markedly increased in sputum (708.7 ? 330.7 pg/ml) and plasma (716.6 ? 162.2 pg/ml) of asthmatic patients and correlated with the percentage of eosinophils in induced sputum. The concentrations of CXCL8 (817.0 ? 105.2 pg/ml) and TNF-??(308.8 ? 96.1 pg/ml) were higher in sputum of COPD patients and correlated with the percentage of neutrophils in induced sputum. There was also an increase in the concentrations of CXCL8 (43.2 ? 6.8 pg/ml) in sputum of asthmatic patients. In the most severe cases of human poisoning by Tityus serrulatus, pulmonary edema is a frequent ?nding and can be the cause of death. Mast cells can release a range of mediators known to be involved in the development of lung edema following T. serrulatus venom injection. The present work was designed to investigate whether mast cells participated in the acute lung injury induced by T. serrulatus scorpion venom and could, thus, be an intermediate between neuropeptide release and activation of the in?ammatory cascade. To this end, mast cells were depleted using compound 48/80. Pulmonary edema, as assessed by the levels of extravasation of Evans blue dye in the bronchoalveolar lavage and in the left lung, was completely inhibited in compound 48/80-treated animals. Moreover, the number of animals surviving 60 min after injection of venom rose from 20 to 60%. Our results demonstrate an important role for mast cells in the development of lung injury and lethality following the intravenous administration of T. serrulatus venom.

Provenance studies carried out on the soft and flint facies of the Capim River kaolin (Northern Brazil) trace the possible sources of sediments that host the ore. Pb?Pb evaporation geochronology was applied to four predominant morphologic classes of detrital zircons, and the ages obtained were compared to the main age intervals of the rocks surrounding the Capim kaolin district (CKD). Four major plateau ages (2.15, 2.02, 1.87, and 1.51 Ga) were defined for both soft and flint facies, indicating a common source for the kaolin and provenance from NE and SW. The 2.15 and 2.02 Ga ages correlate with the granitic bodies of the Gurupi region, located NE of the study area. The 1.87 and 1.51 Ga ages show provenance from the Amazon Craton, the former from the southern portion, in the Caraja?s region, and the latter from the southwestern portion. Although less abundant, Archean (3.18, 2.79, and 2.55 Ga) and Neoproterozoic ages (0.8 and 0.51 Ga) also correlate with the SW and NE provenances, respectively. The first corresponds to the oldest rocks of the Caraja?s region and the second to the rocks of the Gurupi region, corresponding to the Brasiliano orogeny.

The present work quantifies the erosive processes in the two main substrates (schists?phyllites and granites?gneisses) of the upper Maracuj? Basin in the Quadril?tero Ferr?fero/MG, Brazil, a region of semi-humid tropical climate. Two measuring methods of concentration were used: (i) in situ produced 10Be in quartz veins (surface erosion rates) and (ii) 10Be in fluvial sediments (basin erosion rates). The results confirm that (i) erosion tends to be more aggressive close to the headwaters than in the lower parts of the basin and (ii) the region is now affected by dissection.

Positron annihilation lifetime and Doppler broadening annihilation radiation line shape on the salts Ln(CF3COO)3?xH2O and the adducts Ln(CF3COO)3(3-picNO)?xH2O (Ln=La?Pr, Sm?Lu and Y, and 3-picNO=3picoline-N-oxide, x in the range 0.75?1.33) were performed. Positroniumformation in the adducts, except for the europium complex, was observed. These results are discussed in terms of the electronic properties of the central metal ions.

The compound formed by the copper-Schiff base complex salicylideneserinatecopper(II), [Cu(sal-ser)(H2O)], interacting with -cyclodextrin was prepared, and characterized in the solid state by infrared, UV-visible and EPR spectroscopies, X- ray diffraction, and thermoanalytical techniques. The catalytic activity of this compound, [Cu(sal-ser)CD], in the decomposition of hydrogen peroxide, and in the dismutation of superoxide radicals was also verified, in comparison with the reactivity of the free complex, in aqueous solution. In both cases, a decreasing in the reaction rate was observed for the CD-containing compound. The results of structural characterization, in addition to the substantial differences observed in the catalytic activities of the compounds, are indicative of partial insertion of the copper complex in the cavity of the oligosaccharide.

This study was carried out in the Vargem das Flores basin, which is located in Betim and Contagem, Minas Gerais State. The principal aim of this research was to study soil erosion by gullies, which occurred with heterogeneous concentrations throughout this basin. Field data and the remote sensing products allowed the characterization and mapping: of geology, geomorphology, gully features and landuses evolution in the last decades. This methodology have been allowing the evaluation of the geological risk.

A drenagem ?cida de mina (DAM) constitui um dos grandes problemas ambientais associados ?s atividades mineradoras. Ela se caracteriza basicamente pela eleva??o da acidez no meio e, consequentemente, pela dissolu??o de metais pesados associados ?s matrizes rochosas. O impacto direto da DAM se reflete na degrada??o da qualidade dos corpos h?dricos, com suas conhecidas consequ?ncias. Inserido na avalia??o da gera??o da DAM, o presente trabalho tem como objetivo principal a obten??o do balan?o h?drico na regi?o do bota-fora BF4, um dos maiores respons?veis pela forma??o da DAM na mina de ur?nio Osamu Utsumi, propriedade das Ind?strias Nucleares do Brasil (INB), situada em Caldas MG. A utiliza??o desta ?ferramenta? hidrol?gica serviu como medida preliminar, na tentativa da determina??o dos principais contribuintes h?dricos e na melhoria do entendimento do fluxo aquoso para a gera??o de DAM, no local. Para o balan?o, dados meteorol?gicos fornecidos pela INB e pelo Instituto Nacional de Meteorologia (INMET) foram utilizados e tamb?m vaz?es h?dricas monitoras pela INB. Considerando-se as hip?teses adotadas, os resultados indicam a precipita??o como o principal contribuinte para o volume de DAM gerado na base do BF4, norteando assim poss?veis medidas de remedia??o.
