Fructose-1,6-bisphosphate reduces inflammatory pain-like behaviour in mice: role of adenosine acting on A(1) receptors

Background and purpose: D-Fructose-1,6-bisphosphate (FBP) is an intermediate in the glycolytic pathway, exerting pharmacological actions on inflammation by inhibiting cytokine production or interfering with adenosine production. Here, the possible antinociceptive effect of FBP and its mechanism of action in the carrageenin paw inflammation model in mice were addressed, focusing on the two mechanisms described above. Experimental approach: Mechanical hyperalgesia (decrease in the nociceptive threshold) was evaluated by the electronic pressure-metre test; cytokine levels were measured by elisa and adenosine was determined by high performance liquid chromatography. Key results: Pretreatment of mice with FBP reduced hyperalgesia induced by intraplantar injection of carrageenin (up to 54%), tumour necrosis factor alpha (40%), interleukin-1 beta (46%), CXCL1 (33%), prostaglandin E(2) (41%) or dopamine (55%). However, FBP treatment did not alter carrageenin-induced cytokine (tumour necrosis factor alpha and interleukin-1 beta) or chemokine (CXCL1) production. Temos mostrado que os glicocorticóides endógenos (GE) modulam o rolling e a aderência de neutrófilos in vivo, mediando a expressão de moléculas de adesão no leucócito e no endotélio. Adicionalmente, os GE controlam a maturação neutrofílica na medula e a sua mobilização para o sangue periférico. O presente trabalho visou investigar os mecanismos moleculares e celulares envolvidos na modulação exercida pelos GE neste processo. Utilizando ratos Wistar submetidos à adrenalectomia bilateral, tratados com RU 38486 ou controles (falso-operados, tratados com veículo ou não manipulados), foi demonstrado que: 1) os GE controlam, negativamente, a expressão de L-selectina em neutrófilos circulantes e ICAM-1, VCAM-1, PECAM-1, VAP-1 na célula endotelial e, positivamente, a expressão de L-selectina em granulócitos da medula óssea via seu receptor citosólico (GCR); 2) o mecanismo envolvido no controle dos GE sobre a expressão de L-selectina é independente de ação sobre sua expressão gênica ou da atividade de NFkB, mas dependente da expressão de anexina-A1, como verificado em camundongos knockouts (KO) para esta proteína 3) o controle da expressão de moléculas de adesão endotelial é dependente de ações sobre a expressão gênica...

The exopolysaccharide, Botryosphaeran, produced by the ligninolytic, ascomycetcous fungus Botryosphaeria sp., was isolated from the extracellular fluid by precipitation with ethanol, and purified by gel permeation chromatography to yield a carbohydrate-rich fraction (96%) composed mainly of glucose (98%). Infra-red and C-13 NMR spectroscopy showed that all the glucosidic linkages were in the beta-configuration. Data from methylation analysis and Smith degradation indicated that Botryosphaeran was a (1 --> 3)-beta-(D)-glucan with approx 22% side branching at C-6. The products obtained from partial acid hydrolysis demonstrated that the side branches consisted of single (1 --> 6)-beta-linked glucosyl, and (1 --> 6)-beta-linked gentiobiosyl residues.[GRAPHICS](C) 2003 Elsevier Ltd. In order to evaluate the performance of a 1-h 75-g oral glucose tolerance test (OGTT) for the diagnosis of gestational diabetes mellitus (GDM), a cohort of 4998 women, 20 years or older, without previous diabetes being treated in prenatal care clinics in Brazil answered a questionnaire and performed a 75-g OGTT including fasting, 1-h and 2-h glucose measurements between their 24th and 28th gestational weeks. Pregnancy outcomes were transcribed from medical registries. GDM was defined according to WHO criteria (fasting: ≥126 mg/dL; 2-h value: ≥140 mg/dL) and macrosomia as a birth weight equal to or higher than 4000 g. Areas under the receiver operator characteristic curve (AUC) were compared and diagnostic properties of various cut-off points were evaluated. The AUCs for the prediction of macrosomia were 0.606 (0.572-0.637) for the 1-h and 0.589 (0.557-0.622) for the 2-h plasma glucose test. Similar predictability was demonstrable regarding combined adverse outcomes: 0.582 (0.559-0.604) for the 1-h test and 0.572 (0.549-0.595) for the 2-h test. When the 1-h glucose test was evaluated against a diagnosis of GDM defined by the 2-h glucose test, the AUC was 0.903 (0.886-0.919). A(1) adenosine receptors inhibit adenylate cyclase by activating G(i)/G(o), whereas A(2A) receptors activate G(s). We examined how regions of A(1) and A(2A) receptors regulate coupling to G-proteins by constructing chimaeras in which the third intracellular loops (3ICL or L) and/or the C-termini (or T) were switched. Pertussis toxin (PTX) was used in membrane radioligand binding assays to calculate the fraction of recombinant receptors coupled to G(i)/G(o) and in whole cells to differentially influence agonist-stimulated cAMP accumulation. Switching A(1)/A(2A) 3ICL domains results in receptors that maintain binding selectivity for ligands but are doubly coupled. Receptor chimaeras with an A(1) 3ICL sequence (A(2A)/A(1)L or A(2A)/A(1)LT) respond to agonist stimulation with elevated cAMP despite being coupled predominantly to G(i)/G(o). These chimaeras have basal cAMP levels lower than those of wild-type A(2A) receptors, similar to wild-type A(1) receptors. The A(1) C-terminus modulates the coupling of receptors with A(1) 3ICL such that A(2A)/A(1)LT is better coupled to G(i)/G(o) than A(2A)/A(1)L. The C-terminus has little impact on coupling to receptors containing A(2A) 3ICL sequence. Our results show that the C-terminus sequence selectively facilitates coupling to G(i)/G(o) mediated by A(1) 3ICL and not by other intracellular domains that favour G(i) coupling. Avenacin A-1 is a member of a group of naturally occurring compounds called saponins. It is found in oat plants, where it protects against fungal pathogens. A combined electrical and optical chamber was used to determine the interaction of avenacin A-1 with Montal-Mueller planar lipid bilayers. This system allowed simultaneous measurement of the effect of avenacin A-1 on the fluorescence and lateral diffusion of a fluorescent lipid probe and permeability of the planar lipid bilayer. As expected, cholesterol was required for avenacin A-1-induced bilayer permeabilization. The planar lipid bilayers were also challenged with monodeglucosyl, bis-deglucosyl, and aglycone derivatives of avenacin A-1. The results show that the permeabilizing activity of the native avenacin A-1 was completely abolished after one, two, or all three sugar residues are hydrolyzed (monodeglucosyl, bis-deglucosyl, and aglycone derivatives, respectively). Fluorescence recovery after photobleaching (FRAP) measurements on cholesterol-containing planar lipid bilayers revealed that avenacin A-1 caused a small but significant reduction in the lateral diffusion of the phospholipid probe N-(7-nitrobenzoyl-2-oxa-1,3-diazol-4-yl)-1, 2-dihexadecanoyl-sn-glycero-3-phosphoethanolamine (NBD-PE). 1,1,1-Trichloroethane (1,1,1-TCA) is a common groundwater pollutant as a result of improper disposal and accidental spills. It is often found as a cocontaminant with trichloroethene (TCE) and inhibits some TCE-degrading microorganisms. 1,1,1-TCA removal is therefore required for effective bioremediation of sites contaminated with mixed chlorinated organics. This study characterized MS, a 1,1,1-TCA-degrading, anaerobic, mixed microbial culture derived from a 1,1,1-TCA-contaminated site in the northeastern United States. MS reductively dechlorinated 1,1,1-TCA to 1,1-dichloroethane (1,1-DCA) and then to monochloroethane (CA) but not further. Cloning of bacterial 16S rRNA genes revealed among other organisms the presence of a Dehalobacter sp. and a Desulfovibrio sp., which are both phylogenetically related to known dehalorespiring strains. Monitoring of these populations with species-specific quantitative PCR during degradation of 1,1,1-TCA and 1,1-DCA showed that Dehalobacter proliferated during dechlorination. Dehalobacter growth was dechlorination dependent, whereas Desulfovibrio growth was dechlorination independent. Amyloid β-protein (Aβ) is central to the pathology of Alzheimer’s disease (AD). Of the two predominant Aβ alloforms, Aβ1–40 and Aβ1–42, the latter forms more toxic oligomers. C-terminal fragments (CTFs) of Aβ were recently shown to inhibit Aβ1–42 toxicity in vitro. Here we studied Aβ1–42 assembly in the presence of three effective CTF inhibitors and an ineffective fragment, Aβ21–30. Using a discrete molecular dynamics approach that recently was shown to capture key differences between Aβ1–40 and Aβ1–42 oligomerization, we compared Aβ1–42 oligomer formation in the absence and presence of CTFs or Aβ21–30 and identified structural elements of Aβ1–42 that correlated with Aβ1–42 toxicity. CTFs co-assembled with Aβ1–42 into large heterooligomers containing multiple Aβ1–42 and inhibitor fragments. In contrast, Aβ21–30 co-assembled with Aβ1–42 into heterooligomers containing mostly a single Aβ1–42 and multiple Aβ21–30 fragments. Amyloid β-protein (Aβ) deposits in brains of Alzheimer’s disease (AD) patients generate proinflammatory cytokines and chemokines that recruit microglial cells to phagocytose Aβ. Nucleotides released from apoptotic cells activate P2Y2 receptors (P2Y2Rs) in macrophages to promote clearance of dead cells. In this study, we investigated the role of P2Y2Rs in the phagocytosis and clearance of Aβ. Treatment of mouse primary microglial cells with fibrillar (fAβ1–42) and oligomeric (oAβ1–42)Aβ1–42 aggregation solutions caused a rapid release of ATP (maximum after 10 min). Furthermore, fAβ1–42 and oAβ1–42 treatment for 24 h caused an increase in P2Y2R gene expression. Treatment with fAβ1–42 and oAβ1–42 aggregation solutions increased the motility of neighboring microglial cells, a response inhibited by pre-treatment with apyrase, an enzyme that hydrolyzes nucleotides. The P2Y2R agonists ATP and UTP caused significant uptake of Aβ1–42 by microglial cells within 30 min, which reached a maximum within 1 h, but did not increase Aβ1–42 uptake by primary microglial cells isolated from P2Y2R−/− mice. BDNF mediates improvements in executive function following a 1-year exercise intervention

Executive function declines with age, but engaging in aerobic exercise may attenuate decline. One mechanism by which aerobic exercise may preserve executive function is through the up-regulation of brain-derived neurotropic factor (BDNF), which also declines with age. The present study examined BDNF as a mediator of the effects of a 1-year walking intervention on executive function in 90 older adults (mean age = 66.82). Participants were randomized to a stretching and toning control group or a moderate intensity walking intervention group. BDNF serum levels and performance on a task-switching paradigm were collected at baseline and follow-up. We found that age moderated the effect of intervention group on changes in BDNF levels, with those in the highest age quartile showing the greatest increase in BDNF after 1-year of moderate intensity walking exercise (p = 0.036). The mediation analyses revealed that BDNF mediated the effect of the intervention on task-switch accuracy, but did so as a function of age, such that exercise-induced changes in BDNF mediated the effect of exercise on task-switch performance only for individuals over the age of 71. Este art??culo sintetiza los principales resultados de una investigaci??n1, realizada en la Comunidad de Madrid, sobre el modelo 1 a 1 en las aulas de Educaci??n Secundaria en relaci??n con la inclusi??n que procura la ???pr??ctica docente??? desarrollada en el contexto de aula. Se han analizado aulas de siete centros de Educaci??n Secundaria: seis centros p??blicos que han participado en el Programa de Institutos de Innovaci??n Tecnol??gica y un centro privado concertado en el que se hace un uso masivo de las TIC, de los 77 casos/aulas distribuidos por varias comunidades aut??nomas de Espa??a. Para ello se ha utilizado metodolog??a cualitativa. Se han dise??ado y empleado como instrumentos el diario del investigador en el que se recogen las observaciones del aula, entrevistas a los coordinadores TIC, entrevistas a los profesores participantes y un cuestionario para el alumnado. Se ha realizado tambi??n un an??lisis documental. Los resultados del estudio revelan que la utilizaci??n de las TIC, y en concreto del aula digital, condiciona los agrupamientos del alumnado y el tipo de metodolog??a practicada. Introducci??n: La probabilidad de padecer un tumor vesical a lo largo de la vida es de m??s del 4% en hombres y de alrededor del 1,2% en mujeres, incidencia que aumenta con la edad, lo que supone m??s de 100000 nuevos casos anuales de c??ncer vesical en Europa, de los cuales un 30% tendr??n invasi??n muscular. En 2006 provoc?? el 4,1 % y 1,8 % del total de muertes por c??ncer en los varones y las mujeres europeos, por lo que el c??lculo del pron??stico es fundamental para optimizar el tratamiento adecuado a cada paciente. Son marcadores de riesgo establecidos la estirpe y el subtipo histol??gico junto con el grado y el estadio tumoral, al igual que la invasi??n linfovascular. Hemos realizado un estudio para valorar la relaci??n existente entre la edad y dichos marcadores de riesgo con la mortalidad. Material y M??todos: Se ha realizado un estudio retrospectivo observacional, incluyendo los pacientes que se realizaron cistectom??a por tumor infiltrante en el Hospital Universitario San Cecilio de Granada (Espa??a) desde enero de 2000 hasta diciembre de 2011. Se recogieron datos sobre caracter??sticas histol??gicas de la pieza quir??rgica. Resultados: De las 172 cistectom??as realizadas durante ese periodo, el 75% se incluyeron en el estudio (129 pacientes) por encontrar en la pieza tumor transicional. 1 CD-ROM : il., mapas; Desde 1.932 se est?? discutiendo acerca de la estratigraf??a y la composici??n de los dep??sitos y rocas que constituyen la Formaci??n Popay??n, surgiendo varias propuestas estratigr??ficas que se han venido actualizando de acuerdo a la evoluci??n de los conceptos que, con el transcurrir del tiempo, ha tenido la vulcanolog??a. A partir de la caracterizaci??n de los dep??sitos de la Formaci??n Popay??n se plantea que est?? constituida primordialmente por dep??sitos de flujos de ignimbritas y por espesos dep??sitos de flujos de ceniza y bloques de hasta 200 m de espesor que rellenaron un paleorelieve, construido sobre rocas del Paleozoico al Paleoceno, conformando la Meseta de Popay??n. Se propone que la secci??n estratigr??fica tipo de la Formaci??n Popay??n sea la levantada en la quebrada Sombrerillo, localizada al norte de la Meseta de Popay??n en el Municipio de Morales, de muy f??cil acceso, y donde est?? perfectamente representada ??sta formaci??n. Las columnas estratigr??ficas levantadas en la Chorrera del Club Campestre del municipio de Popay??n y en la quebrada La Chorrera- r??o Hondo, al sur de la meseta, en el municipio de El Tambo, se consideran secciones estratigr??ficas de referencia. En la secci??n estratigr??fica de la quebrada Sombrerillo se observa la base de la Formaci??n Popay??n conformada por dep??sitos de flujos de ignimbritas con un espesor m??ximo observado de 100 m; estas ignimbritas est??n suprayacidas por una serie de dep??sitos de flujos pirocl??sticos y dep??sitos vulcano sedimentarios...

Este artículo narra la experiencia de un proyecto piloto de formación docente en servicio en el marco del modelo denominado 1 a 1 durante el año 2011, que destina un ordenador para cada estudiante de escuelas secundarias públicas de la República Argentina. Se plantea que la formación y el acompañamiento de los docentes en la implementación del modelo 1 a 1 deben ser entendidos como formación en servicio y de carácter institucional, dada la importancia de considerar la escuela como un caso en sí mismo, aunque obedezca a políticas públicas generales a todos los centros de secundaria. De esta experiencia, surgieron algunos rasgos a considerar: la importancia del equipo directivo; la validación de las redes sociales en cuanto a nuevos formatos comunicativos; los estudiantes como propios agentes del cambio en las prácticas áulicas, y el lugar de la emoción en las innovaciones de este tipo, donde cambian los perfiles identitarios tradicionales de la comunidad educativa.; ; Aquest article narra l’experiència d’un projecte pilot de formació docent en servei en el marc del model denominat 1 a 1 durant l’any 2011, que destina un ordinador per a cada estudiant d’escoles secundàries públiques de la República Argentina. Con la entrada en vigencia de la Resolución 0627 de 2006, reglamentándose la altura de 4m sobre el nivel del piso como estrategia de medición para evaluar los niveles de ruido ambiental, se ha creado una gran controversia entre las entidades ambientales gubernamentales, los académicos y profesionales del sector, sobre las implicaciones económicas, logísticas y operativas para el desarrollo del trabajo de campo. Con el propósito de determinar si mediciones simultáneas de ruido urbano efectuadas a 1,5 m y 4 m representan o no la misma realidad sonora, se realizó un análisis estadístico a un conjunto de datos obtenidos en el marco de la construcción del mapa de ruido del municipio. Después de un análisis de los datos agrupados por punto, por jornada (diurna y nocturna) y por tipo de día (hábil y no hábil), se infiere que no es posible afirmar la semejanza entre muestras de ruido tomadas a 1,5 m y 4 m de altura, no sólo por las diferencias numéricas, sino que representan realidades sonoras diferentes.
