This PhD thesis presents the development of a prosody system for European Portuguese (EP) for text-to-speech (TTS) applications. Basically, TTS systems carry out the automatic utterance of a text and consist in a sequence of several modules. Those modules implement the pre-processing of the text input, the phonetic transcription and the supra-segmental processing that consists in the inclusion of prosodic patterns. The prosody is responsible for a communicative intention and guarantees some naturalness in the uttered speech. The prosodic features consist in the imposition of the timing, characterized by the segmental durations and pauses, the intonation, characterized by the fundamental frequency (F0) curve, and by the intensity curve. The preparatory work that was fundamental for modelling and testing purposes is presented in the beginning. It starts with a preliminary study about the stressed syllable. This study identifies the variation range of F0, duration and intensity features in stressed syllable along contexts. Then the FEUP-IPB EP speech database that was used in following studies is presented. The database is labelled at the levels of the phoneme, word, sentence and F0. The thesis follows on with the presentation of two algorithms to provide the syllabic splitting of the text and of the phoneme sequences. No presente trabalho investigamos a relevância dos mediadores lipídicos (Leucotrienos) gerados pela enzima 5-Lipoxigenase (5-LO) na susceptibilidade ou resistência de camundongos ao Melanoma experimental com células tumorais B16-F0, utilizando como modelo camundongos produtores de leucotrienos (129_WT) e camundongos geneticamente deficientes "knockout" de 5-LO (129_5-LO KO). Primeiramente, verificamos que leucócitos peritoneais provenientes de animais WT implantados com melanoma B16-F0, apresentam aumento da expressão do gene para 5-LO (Alox5). Nossos resultados mostram que animais 5-LO KO, deficientes de 5-LO são mais eficientes no controle da progressão do tumor e apresentam significativo aumento na sobrevivência, quando comparados a animais WT, produtores de 5-LO. A nossa análise do perfil imunológico em células esplênicas indicam que a maior eficiência dos camundongos 5-LO KO no controle do crescimento de células tumorais B16-F0 estariam associados à presença numérica aumentada de neutrófilos (Gr-1+), células apresentadoras de antígeno (I-Ab+) majoritariamente CD19+CD80+ e esplenócitos capacitados para produção de altos níveis de citocinas pró-inflamatórias/efetoras como a IL-6, TNF?, IFN-? e baixos níveis de citocinas regulatórias como IL-10...

To evaluate whether expression of cloned genes for the F0 proton channel of the Escherichia coli F1F0 ATPase is sufficient to cause membrane proton permeability, plasmids carrying different combinations of the uncB, E, and F genes, encoding the a, c, and b subunits of the F0 sector, cloned behind the inducible lac promoter in pUC9 or pUC18, were constructed. The effects of inducing F0 synthesis in an unc deletion strain were monitored by measuring cell growth rate, quantitating F0 subunits by immunoblotting, and measuring the ability of membranes to maintain a respiration-induced proton gradient and to bind F1 and carry out energy-coupling reactions. The levels of functional reconstitutable F0 in membranes could be increased four- to sixfold with no change in cellular growth rate or membrane proton permeability (assayed by fluorescence quenching). These results were obtained in uninduced cultures, so the F0 genes were presumably being transcribed from some promoter besides lac. Induction of transcription of all three F0 genes produced increased amounts of F0 subunits in membranes as determined by immunoblot and F1-binding assays, but, when reconstituted with F1, the F0 in membranes isolated from induced cultures was significantly less functional than the F0 in membranes isolated from uninduced cultures. During the assembly of the Escherichia coli proton-translocating ATPase, the subunits of F1 interact with F0 to increase the proton permeability of the transmembrane proton channel. We tested the involvement of the delta subunit in this process by partially and completely deleting uncH (delta subunit) from a plasmid carrying the genes for the F0 subunits and delta and testing the effects of those F0 plasmids on the growth of unc+ and unc mutant E. coli strains. We found that the delta subunit was required for inhibition of growth of unc+ cells. We also tested membranes isolated from unc-deleted cells containing F0 plasmids for F1-binding ability. In unc-deleted cells, these plasmids produced F0 in amounts comparable to those found in normal unc+ E. coli cells, while having only small effects on cell growth. These studies demonstrate that the delta subunit plays an important role in opening the F0 proton channel but that it does not serve as a temporary plug of F0 during assembly, as had been previously speculated (S. Pati and W. S. A. Brusilow, J. Biol. Chem. 264:2640-2644, 1989).

The uncB, E, F, and H genes of the Escherichia coli unc operon were cloned behind the lac promoter of plasmid pUC9, generating plasmid pBP101. These unc loci code, respectively, for the chi, omega, and psi subunits of the F0 sector and the delta subunit of the F1 sector of the H+-ATP synthase complex. Induction of expression of the four unc genes by the addition of isopropyl-beta-D-thiogalactoside resulted in inhibition of growth. During isopropyl-beta-D-thiogalactoside induction, the three subunits of F0 were integrated into the cytoplasmic membrane with a resultant increase in H+ permeability. A functional F0 was formed from plasmid pBP101 in a genetic background lacking all eight of the unc structural genes coding the F1F0 complex. In the unc deletion background, a reasonable correlation was observed between the amount of F0 incorporated into the membrane and the function measured, i.e., high-affinity binding of F1 and rate of F0-mediated H+ translocation. This correlation indicates that most or all of the F0 assembled in the membrane is active. Although the F0 assembled under these conditions binds F1, only partial restoration of NADH-dependent or ATP-dependent quenching of quinacrine fluorescence was observed with these membranes. Proteolysis of a fraction of the psi subunit may account for this partial deficiency. A strain of Escherichia coli (AN1007) carrying the polar uncD436 allele which affects the operon coding for the F1-F0 adenosine triphosphatase (ATPase) complex was isolated and characterized. The uncD436 allele affected the two genes most distal to the operon promoter, i.e., uncD and uncC. Although the genes coding for the F0 portion of the ATPase complex were not affected in strains carrying this mutant allele, the lack of reconstitution of washed membranes by normal F1 ATPase suggested that a functional F0 might not be formed. This conclusion was supported by the observation that the 18,000-molecular-weight F0 subunit, coded for by the uncF gene, was absent from the membranes. Plasmid pAN36 (uncD+C+), when inserted into a strain carrying the uncD436 allele, resulted in the incorporation of the 18,000-molecular-weight F0 subunit into the membrane. A further series of experiments with Mu-induced polarity mutants, with and without plasmid pAN36, showed that the formation of both the alpha- and beta-subunits of F1 ATPase was an essential prerequisite to the incorporation into the membrane of the 18,000-molecular-weight F0 subunit and to the formation of a functional F0. Examination of the polypeptide composition of membranes from various unc mutants allowed a sequence for the normal assembly of the F1-F0 ATPase complex to be proposed.

Archae possess unique biochemical systems quite distinct from the pathways present in eukaryotes and eubacteria. 7,8-Dimethyl-8-hydroxy-5deazaflavin (F0) and F420 are unique deazaflavin-containing coenzyme and methanogenic signature molecules, essential for a variety of biochemical transformations associated with methane biosynthesis and light-dependent DNA repair. The deazaflavin cofactor system functions during methane biosynthesis as a low-potential hydrid shuttle F420/F420H2. In DNA photolyase repair proteins, the deazaflavin cofactor is in the deprotonated state active as a light-collecting energy transfer pigment. As such, it converts blue sunlight into energy used by the proteins to drive an essential repair process. Analysis of a eukaryotic (6-4) DNA photolyase from Drosophila melanogaster revealed a binding pocket, which tightly binds F0. Residues in the pocket activate the cofactor by deprotonation so that light absorption and energy transfer are switched on. The crystal structure of F0 in complex with the D. melanogaster protein shows the atomic details of F0 binding and activation, allowing characterization of the residues involved in F0 activation. The membrane-bound component F0, which is a major component of the F0F1-ATP synthase, works as a rotary motor and plays a central role in driving the F1 component to transform chemiosmotic energy into ATP synthesis. We conducted molecular dynamics simulations of b2-free F0 in a 1-palmitoyl-2-oleoyl-phosphatidylcholine lipid bilayer for tens of nanoseconds with two different protonation states of the cAsp-61 residue at the interface of the a-c complex in the absence of electric fields and under electric fields of ±0.03 V/nm across the membrane. To our surprise, we observed that the upper half of the N-terminal helix of the c1 subunit rotated about its axis clockwise by 30°. An energetic analysis revealed that the electrostatic repulsion between this N-terminal helix and subunit c12 was a major contributor to the observed rotation. A correlation map analysis indicated that the correlated motions of residues in the interface of the a-c complex were significantly reduced by external electric fields. The deuterium order parameter (SCD) profile calculated by averaging all the lipids in the F0-bound bilayer was not very different from that of the pure bilayer system, in agreement with recent 2H solid-state NMR experiments. In this study we investigated grouping-related F0 patterns in Mandarin by examining the effect of syllable position in a group while controlling for tone, speaking mode, number of syllables in a group, and group position in a sentence. We analyzed syllable duration, F0 displacement, ratio of peak velocity to F0 displacement (vp/d ratio) and shape of F0 velocity profile (parameter C) in sequences of Rising, Falling and High tones. Results showed that syllable duration had the most consistent grouping-related patterns. In a short phrase of 1–4 syllables, duration is longest in the final position, second longest in the initial position, and shortest in the medial positions. In Rising and Falling tone sequences, syllable duration was positively related to F0 displacement, but negatively related to vp/d ratio. Sequences consisting of only the High tone, however, showed no duration-matching F0 variations. Modeling simulations with a second-order linear system showed that duration variations alone could generate F0 displacement and vp/d ratio variations comparable to those in actual data. We interpret the results as evidence that grouping is encoded directly by syllable duration, while the corresponding variations in F0 displacement, vp/d ratio and velocity profile are the consequences of duration control.

Do Red Deer Stags (Cervus elaphus) Use Roar Fundamental Frequency (F0) to Assess Rivals?

It is well established that in humans, male voices are disproportionately lower pitched than female voices, and recent studies suggest that this dimorphism in fundamental frequency (F0) results from both intrasexual (male competition) and intersexual (female mate choice) selection for lower pitched voices in men. However, comparative investigations indicate that sexual dimorphism in F0 is not universal in terrestrial mammals. In the highly polygynous and sexually dimorphic Scottish red deer Cervus elaphus scoticus, more successful males give sexually-selected calls (roars) with higher minimum F0s, suggesting that high, rather than low F0s advertise quality in this subspecies. While playback experiments demonstrated that oestrous females prefer higher pitched roars, the potential role of roar F0 in male competition remains untested. Here we examined the response of rutting red deer stags to playbacks of re-synthesized male roars with different median F0s. Our results show that stags’ responses (latencies and durations of attention, vocal and approach responses) were not affected by the F0 of the roar. This suggests that intrasexual selection is unlikely to strongly influence the evolution of roar F0 in Scottish red deer stags, and illustrates how the F0 of terrestrial mammal vocal sexual signals may be subject to different selection pressures across species. Further investigations on species characterized by different F0 profiles are needed to provide a comparative background for evolutionary interpretations of sex differences in mammalian vocalizations.

Recent psychophysical studies suggest that normal-hearing (NH) listeners can use acoustic temporal-fine-structure (TFS) cues for accurately discriminating shifts in the fundamental frequency (F0) of complex tones, or equal shifts in all component frequencies, even when the components are peripherally unresolved. The present study quantified both envelope (ENV) and TFS cues in single auditory-nerve (AN) fiber responses (henceforth referred to as neural ENV and TFS cues) from NH chinchillas in response to harmonic and inharmonic complex tones similar to those used in recent psychophysical studies. The lowest component in the tone complex (i.e., harmonic rank N) was systematically varied from 2 to 20 to produce various resolvability conditions in chinchillas (partially resolved to completely unresolved). Neural responses to different pairs of TEST (F0 or frequency shifted) and standard or reference (REF) stimuli were used to compute shuffled cross-correlograms, from which cross-correlation coefficients representing the degree of similarity between responses were derived separately for TFS and ENV. For a given F0 shift, the dissimilarity (TEST vs. REF) was greater for neural TFS than ENV. A phenomenological analysis of the scalar meson f0(980) is performed that relies on the quasi-two body decays D and Ds -> f0(980)P, with P=pi, K. The two-body branching ratios are deduced from experimental data on D or Ds -> pi pi pi, K Kbar pi and from the f0(980) -> pi+ pi- and f0(980) -> K+ K- branching fractions. Within a covariant quark model, the scalar form factors F0(q2) for the transitions D and Ds -> f0(980) are computed. The weak D decay amplitudes, in which these form factors enter, are obtained in the naive factorization approach assuming a quark-antiquark state for the scalar and pseudoscalar mesons. They allow to extract information on the f0(980) wave function in terms of u-ubar, d-dbar and s-sbar pairs as well as on the mixing angle between the strange and non-strange components. The weak transition form factors are modeled by the one-loop triangular diagram using two different relativistic approaches: covariant light-front dynamics and dispersion relations. We use the information found on the f0(980) structure to evaluate the scalar and vector form factors in the transitions D and Ds -> f0(980), as well as to make predictions for B and Bs -> f0(980), for the entire kinematically allowed momentum range of q2.; Comment: 45 pages...

Within a nonlinear chiral Lagrangian framework the correlations between the quark and glueball admixtures of the isosinglet scalar mesons below 2 GeV and the current large uncertainties on the mass of the f0(600) and the f0(1370) are studied. The framework is formulated in terms of two scalar meson nonets (a two-quark nonet and a four-quark nonet) together with a scalar glueball. It is shown that while some properties of these states are sensitive to the mass of f0(600) and f0(1370), several relatively robust conclusions can be made: The f0(600), the f0(980), and the f0(1370) are admixtures of two and four quark components, with f0(600) being dominantly a non-strange four-quark state, and f0(980) and f0(1370) having a dominant two-quark component. Similarly, the f0(1500) and the f0(1710) have considerable two and four quark admixtures, but in addition have a large glueball component. For each state, a detailed analysis providing the numerical estimates of all components is given. It is also shown that this framework clearly favors the experimental values: m[f0(600)] < 700 MeV and m[f0(1370)] = 1300-1450 MeV. Moreover, an overall fit to the available data shows a reciprocal substructure for the f0(600) and the f0(1370), and a linear correlation between their masses of the form m [f0(1370)] = 0.29 m[f0(600)] + 1.22 GeV. In the article of V.V. Anisovich et al., Yad. Fiz. 63, 1489 (2000), the K-matrix solutions for the wave IJ^{PC}=00^{++} were obtained in the mass region 450 - 1900 MeV where four resonances f0(980), f0(1300), f0(1500), f0(1750) and the broad state f0(1530^{+90}_{-250}) are located. Various processes have been proposed previously to study a0(980)-f0(980) mixing through f0(980)->a0(980) transition. Here we investigate in detail the difference between a0(980)->f0(980) and f0(980)->a0(980) transitions. It is found that the a0(980)->f0(980) transition can provide additional constrains to the parameters of a0(980) and f0(980) mesons. Using the U(3)R x U(3)L extended Linear Sigma Model with the ordinary (pseudo)scalar and (axial)vector mesons as well as a scalar glueball, we study the vacuum phenomenology of the scalar-isoscalar resonances f0(1370), f0(1500) and f0(1710). Using e+e- -> Ds^-Ds^{*+} and Ds^{*-}Ds^+ interactions at 4170 MeV collected with the CLEO-c detector, we investigate the semileptonic decays Ds+ -> f0(980) e+ nu, and Ds+ -> phi e+ nu. By examining the decay rates as functions of the four-momentum transfer squared, q^2, we measure the ratio [dB/dq^2(Ds+ -> f0(980) e+ nu) B(f0 -> pi+pi-)]/[dB/dq^2(Ds+ -> phi e+ nu)B(phi -> K+K-)] at q^2 of zero to be (42 +/- 11)%. This ratio has been predicted to equal the rate ratio [B(Bs -> J/psi f_0)B(f0 -> pi+pi-)]/[B(Bs -> J/psi phi)B(phi -> K+K-)], thus indicating that the CP eigenstate J/psi f0 could be useful for measuring CP violation via Bs mixing. Assuming a simple pole model for the form factor |f+(q^2)| in the f0 e+ nu decay, we find a pole mass of (1.7 {+4.5} {-0.7} +/- 0.2) GeV. We also determine the f0 mass and width as (977 {+11}{-9} +/- 1) MeV, and (91{+30}{-22}+/- 3) MeV, respectively. In addition, we present updated results for B(Ds+ -> f0(980) e+ nu) B(f0 -> pi+pi-)=(0.20 +/- 0.03 +/- 0.01)%, and B}(Ds+ -> phi e+ nu)=(2.36 +/- 0.23 +/- 0.13)%.; Comment: 19 pages, 9 figures, to be published in Physical Review D

For the first time a complete data set of the two-body decays of the f0(1370), f0(1500) and f0(1710) into all pseudoscalar mesons is available. The implications of these data for the flavour content for these three f0 states is studied. For the first time a complete data set of the two-body decays of the f0(1370), f0(1500) and f0(1710) into all pseudoscalar mesons is available. The implications of these data for the flavour content for these three f0 states is studied. The production and decay properties of the f0(1370), f0(1500), f0(2000) and f2(1950) have been studied in central pp interactions at 450 GeV/c. The dPT, phi and |t| distributions of these resonances are presented. For the J = 0 states, the f0(1370) and f0(2000) have similar dPT and phi dependences. These are different to the dPT and phi dependences of the f0(980), f0(1500) and f0(1710). For the J = 2 states the f2(1950) has different dependences to the f2(1270) and f2'(1520). This shows that the dPT and phi dependences are not just J phenomena.; Comment: 14 pages, Latex, 4 Figures
