In silico analysis of microRNAS targeting the HLA-G 3 ` untranslated region alleles and haplotypes

It has been reported that microRNAs (miRNA) may have allele-specific targeting for the 3` untranslated region (3` UTR) of the HLA-G locus. In a previous study, we reported 11 3`UTR haplotypes encompassing the 14-bp insertion/deletion polymorphism and seven SNPs (+3003 T/C, +3010 C/G, +3027 C/A, +3035 C/T, +3142 C/G, +3187A/G,and +3196 C/G), of which only the +3142 C/G SNP has been reported to influence the binding of miRNAs. Using bioinformatics analyses, we identified putative miRNA-binding sites considering the haplotypes encompassing these eight polymorphic sites, and we ranked the lowest free energies that could potentially lead to an mRNA degradation or translational repression. When a specific haplotype or a particular SNP was associated with a miRNA-binding site, we defined a free energy difference of 4 kcal/mol between alleles to classify them energetically distant. The best results were obtained for the miR-513a-5p, miR-518c*, miR-1262 and miR-92a-1*, miR-92a-2*, miR-661, miR-1224-5p, and miR-433 miRNAs, all influencing one or more of the +3003, +3010, +3027, and +3035 SNPs. The HLA-G gene is predominantly expressed at the maternal-fetal interface. It has been associated with maternal-fetal tolerance and in the inhibition of cytotoxic T lymphocyte and natural killer cytolytic functions. At least two variations in the 3` untranslated region (UTR) of HLA-G locus are associated with HLA-G expression levels, the 14-bp deletion/insertion polymorphism and the +3142 single-nucleotide polymorphism (SNP). However, this region has not been completely characterized yet. The variability of the 3`UTR of HLA-G gene and its haplotype structure were characterized in 155 individuals from Brazil, as well as HLA-G alleles associated with each of the 3`UTR haplotype. The following eight variation sites were detected: the 14-bp polymorphism and SNPs at the positions +3003T/C, +3010C/G, +3027A/C, +3035C/T, +3142G/C, +3187A/G and +3196C/G. Similarly, 11 different 3`UTR haplotypes were identified and several HLA-G alleles presented only one 3`UTR haplotype. In addition, a high linkage disequilibrium among the variation sites was detected, especially among the 14-bp insertion and the alleles +3142G and +3187A, all previously associated with low mRNA availability, demonstrating that their effects are not independent. Objetivo: O objetivo desse estudo foi identificar a associação entre fatores clínicos e laboratoriais com o uso de insulina em gestantes com DMG no momento do diagnóstico e analisar os possíveis fatores preditores do uso de insulina. Método: Foram estudadas, de forma retrospectiva, 294 pacientes com diabetes melito gestacional (DMG) diagnosticado por meio do teste de tolerância à glicose oral de 100 gramas (TTGO-100g) entre 24 e 33 semanas completas de gestação, cujo seguimento pré-natal foi realizado ambulatorialmente pelo setor de Endocrinopatias e Gestação da Clínica Obstétrica do Hospital das Clínicas da Faculdade de Medicina da Universidade de São Paulo, no período de 1 de julho de 2002 a 30 de junho de 2008. The aim of this research work was to investigate the influence of fish oil supplementation on the concentration of conjugated linoleic acid (CLA) and omega 6 and 3 in samples of buffalo milk. A total of 24 female buffaloes separated at random into three groups were fed for 49 days with: natural pasture (group I), supplemented with 70mL of fish oil (group II) and 140mL of fish oil (group III). In the experiment the concentration of CLA showed differences (P<0.05) among the three groups, with a maximum of 7.14mg/g fat in group II. No significant differences were found in omega-6 among the three groups. The highest value of 3.82mg/g fat corresponded to group I, whicht had not been supplemented with fish oil. Significant differences were observed in omega 3 (P<0.05) in groups II and III with respect to group I. The highest average value of 2.42mg/g fat was obtained in group III. The closest relationship omega 6/3 (1.37:1) was observed in group III. As a result, the diets of groups II and III, which included fish oil, increased significantly the content of CLA and omega 3 with reductions in levels of omega 6.

Níveis intracelulares de G-3-PDH (sn-glicerol-3-fosfato:NAD+ 2oxidoredutase, EC 1.1.1.8) de levedura de panificação foram acompanhados durante a estocagem sob três diferentes temperaturas. Semelhantes valores de biomassa final e de atividade específica da enzima foram obtidos após crescimento por 48 horas de duas linhagens de leveduras de panificação. O melhor meio (meio indutor) para obtenção de G-3-PDH foi: extrato de levedura (1%,p/v), peptona (2%,p/v), glicerol (3%,v/v) e etanol (1%,v/v). O choque osmótico com adição de NaCl 0,6 M provocou aumento da atividade de G-3-PDH de 1,2 vezes para leveduras crescidas em meio indutor por 48 horas e transferidas para o meio salino, por 2 horas. A estocagem (até 10 dias) da linhagem de levedura GD0, sob temperatura ambiente (27 ºC) estimulou a síntese da G-3-PDH de células propagadas no meio indutor, lavadas e liofilizadas. Estocagens em geladeira (temperatura de 4 - 5 ºC) ou em 'freezer' (temperatura de -18 ºC) mantiveram a atividade da G-3-PDH por até 8 meses.

Heterotrimeric G proteins are comprised of a guanine nucleotide binding Gα subunit and the Gβγ dimers that link G protein-coupled receptors (GPCRs) to effectors. This study focuses on the expression and localization patterns for certain Gβ and Gγ subunits in neonatal and adult cardiomyocytes. We identify developmental downregulation of Gβ1, Gβ2 and Gγ2, and a switch in the molecular form of Gγ3, in cardiomyocytes. Gβ1 is highly localized to caveolae membranes, whereas Gβ2 is identified in caveolae and other membrane fractions. Gβ3 is not detected in neonatal cardiomyocytes, but rather Gβ3 is upregulated in adult cardiomyocytes and detected in the caveolae and soluble fractions. The observation that cardiomyocytes co-express multiple Gβ and Gγ subunits in a developmentally regulated manner, and that these Gβ and Gγ subunits assume distinct subcellular localization patterns, provides for a high level of signaling specificity in the heart.

Heterotrimeric G-proteins, comprising Gα and Gβγ subunits, couple metabotropic receptors to various downstream effectors and contribute to assembling and trafficking receptor based signaling complexes. A G-protein β-subunit, Gβ3 plays a critical role in several physiological processes as a polymorphism in its gene is associated with a risk factor for several disorders. Retinal ON bipolar cells express Gβ3, and they provide an excellent system to study its role. In the ON bipolar cells, mGluR6 inverts the photoreceptor’s signal via a cascade in which glutamate released from photoreceptors closes the TRPM1 channel. This cascade is essential for vision since deficiencies in its proteins lead to complete congenital stationary night blindness. Here we report that Gβ3 participates in the G-protein heterotrimer that couples mGluR6 to TRPM1. Gβ3 deletion in mouse greatly reduces the light response under both scotopic and photopic conditions, but it does not eliminate it. In addition, Gβ3 deletion causes mislocalization and downregulation of most cascade elements and modulators. Furthermore, Gβ3 may play a role in synaptic maintenance since in its absence, the number of invaginating rod bipolar dendrites is greatly reduced, a deficit that was not observed at 3 weeks...

A spice variant of Gβ3, termed Gβ3s, has been associated with the C825T polymorphism in the Gβ3 gene and linked with many human disorders. However, the biochemical properties and functionality of Gβ3s remain controversial. Here, using multidisciplinary approaches including co-immunoprecipitation analysis and bioluminescence resonance energy transfer (BRET) measurements, we showed that unlike Gβ3, Gβ3s failed to form complexes with either Gγ or Gα subunits. Moreover, using a mutant Gγ2 deficient in lipid modification to purify Gβ3s from Sf9 cells without the use of detergents, we further showed that the failure of Gβ3s to form dimers with Gγ was not due to the instability of the dimers in detergents, but rather, reflected the intrinsic properties of Gβ3s. Additional studies indicated that Gβ3s is unstable, and unable to localize properly to the plasma membrane and to activate diverse Gβγ effectors including PLCβ2/3, PI3Kγ, ERKs and the Rho guanine exchange factor (RhoGEF) PLEKHG2. Thus, these data suggest that the pathological effects of Gβ3 C825T polymorphism may result from the downregulation of Gβ3 function. Both n-6 and n-3 polyunsaturated fatty acids (PUFA) are recognized as essential nutrients in the human diet, yet reliable data on population intakes are limited. The aim of the present study was to ascertain the dietary intakes and food sources of individual n-6 and n-3 PUFA in the Australian population. An existing database with fatty acid composition data on 1690 foods was updated with newly validated data on 150 foods to estimate the fatty acid content of foods recorded as eaten by 10,851 adults in the 1995 Australian National Nutrition Survey. Average daily intakes of linoleic (LA), arachidonic (AA), alpha-linolenic (LNA), eicosapentaenoic (EPA), docosapentaenoic (DPA), and docosahexaenoic (DHA) acids were 10.8, 0.052, 1.17, 0.056, 0.026, and 0.106 g, respectively, with long-chain (LC) n-3 PUFA (addition of EPA, DPA, and DHA) totaling 0.189 g; median intakes were considerably lower (9.0 g LA, 0.024 g AA, 0.95 g LNA, 0.008 g EPA, 0.006 g DPA, 0.015 g DHA, and 0.029 g LC n-3 PUFA). OBJECTIVE: To determine the impacts of induced parturition, birth weight, birth order, litter size, and sow parity on piglet serum concentrations of immunoglobulin G (IgG). MATERIALS AND METHODS: In Experiment 1, sows were either induced to farrow (n = 56) or allowed to farrow naturally (n = 84). Litters of induced sows were placed immediately into a warm crèche until farrowing was complete, then all piglets were weighed and placed with the sow at the same time. Blood samples were collected at 3 days of age from one or two of the smallest pigs, one medium pig, and the largest pig in each litter for measurement of serum IgG. In Experiment 2, the firstborn and last-born piglets in 78 litters were blood sampled at 3 days of age and sera were assayed for total IgG. RESULTS: Experiment 1. Mean serum IgG concentration was higher in piglets from induced litters than in piglets from control sows (P < .001). Serum IgG concentrations increased with increased piglet weight (P < .001). The non-thermal nature of the X-ray emission from the shell-type supernova remnants (SNRs) G1.9+0.3 and G330.2+1.0 is an indication of intense particle acceleration in the shock fronts of both objects. This suggests that the SNRs are prime candidates for very-high-energy (VHE; E > 0.1 TeV) γ-ray observations. G1.9+0.3, recently established as the youngest known SNR in the Galaxy, also offers a unique opportunity to study the earliest stages of SNR evolution in the VHE domain. The purpose of this work is to probe the level of VHE γ-ray emission from both SNRs and use this to constrain their physical properties. Observations were conducted with the H.E.S.S. (High Energy Stereoscopic System) Cherenkov Telescope Array over a more than six-year period spanning 2004–2010. The obtained data have effective livetimes of 67 h for G1.9+0.3 and 16 h for G330.2+1.0. The data are analysed in the context of the multiwavelength observations currently available and in the framework of both leptonic and hadronic particle acceleration scenarios. No significant γ-ray signal from G1.9+0.3 or G330.2+1.0 was detected. Aberrant activation of β-catenin is a common event in AML and is an independent predictor of poor prognosis. Although increased β-catenin signaling in AML has been associated with oncogenic translocation products and activating mutations in the FLT3R, the mechanisms that activate β-catenin in AML more broadly are still unclear. Here, we describe a novel link between IL-3 signaling and the regulation of β-catenin in myeloid transformation and AML. In a murine model of HoxB8 and IL-3 cooperation, we show that β-catenin protein levels are modulated by IL-3 and that Cre-induced deletion of β-catenin abolishes IL-3-dependent growth and colony formation. In IL-3-dependent leukemic TF-1.8 cells, we observed increased β-catenin protein levels and nuclear localization in response to IL-3, and this correlated with transcriptional induction of β-catenin target genes. Furthermore, IL-3 promoted β-catenin accumulation in a subset of AML patient samples, and gene-expression profiling of these cells revealed induction of WNT/β-catenin and TCF4 gene signatures in an IL-3-dependent manner. Human leukocyte antigen-G (FILA-G) plays a well-recognized role in the modulation of the immune response, and HLA-G expression has been associated with increased graft survival and decreased rejection episodes. To investigate the role of the HLA-G 3' untranslated region (3'UTR) in renal transplantation, we evaluated several polymorphic sites (14-bp Del/Ins +3003T/C, +3010C/G, +3027C/A, +3035C/T, +3142G/C, and +3187A/G) in patients exhibiting or not exhibiting rejection episodes. A total of 104 patients (15 with acute and 48 with chronic rejection, and 41 with no rejection) and 142 healthy individuals were studied. HLA-G 3'UTR was typed by direct sequencing. The +3035C-C genotype was more frequent in patients exhibiting chronic rejection compared with healthy controls, and the +3035C-T genotype was less frequent in chronic rejection compared with patients without rejection (acute plus chronic) or compared with healthy controls. The +3187G-A genotype, in which the A allele is associated with increased mRNA degradation, showed increased frequency in the rejection group (acute plus chronic) when compared with healthy controls. The 14 base pair Deletion/Insertion genotype was marginally increased in patients with acute rejection. The synthesis of alternating hexamers (8-13) derived from d(C-G)3 or d(G-C)3 but containing c7z8Gd (2) or c7Gd (3) instead of dG is described employing phosphoramidite-chemistry. Apart from the isobutyryl group the dimethylaminomethylene residue was used for the nucleobase-protection of 3. The methyl- and the cyanoethyl-phosphoramidites of 3 (5a-c) were synthesized. They were employed together with those of c7G or c7z8Gd in automated oligonucleotide synthesis. Tm-values as well as thermodynamic data of the oligomers 9, 10, 12, and 13 indicated that duplexes were destabilized if c7Gd replaced dG, whereas c7z8Gd stabilized the duplex structure. In contrast to d(C-G)3 which underwent salt-dependent B-Z transition, CD-spectra of oligomers containing c7Gd or c7z8Gd in place of dG showed retained B-conformation.

Inspired by the recent results of the WASA-at-COSY Collaboration, in which they found a resonance pole in the coupled $^{3}D_{3}$ - $^{3}G_{3}$ partial waves as expected from the $d^{*}$ resonance hypothesis, we calculated the resonance structure in the coupled $^{3}D_{3}$ - $^{3}G_{3}$ partial wave phase shifts of nucleon-nucleon scattering in the framework of two constituent quark models: the quark delocalization color screening model and the chiral quark model. Our results show that there is a resonance $^{7}S_{3}^{\Delta\Delta}$ in the coupled $^{3}D_{3}^{NN}$ and $^{3}G_{3}^{NN}$ partial waves in both of these two models, which is in accordance with the expectation from the $d^{*}$ resonance structure. The resonance shape in the $^{3}D_{3}^{NN}$ partial wave is remarkable, whereas in the $^{3}G_{3}^{NN}$ phase shifts there is a small rise around the resonance energy. This result is in agreement with the recent experimental observations of WASA-at-COSY Collaboration.; Comment: 5 pages, 3 figures

By using properties of Markov homogeneous chains and Banach measure in $\mathrm{N}$, it is proved that a relative frequency of even numbers in the sequence of $n$-th coordinates of all Collatz sequences is equal to the number $\frac{2}{3}+\frac{(-1)^{n+1}}{3\times 2^{n+1}}.$ It is shown also that an analogous numerical characteristic for numbers of the form $3m+1$ is equal to the number $\frac{3}{5}+ \frac{(-1)^{n+1}}{15 \times 2^{2(n-1)}}. $ By using these formulas it is proved that under Collatz conjecture the Collatz mapping has no an asymptotic mixing property $\pmod{3}$. It is constructed also an example of a real-valued function on the cartesian product $N^2$ of the set of all natural numbers $N$ such that an equality its repeated integrals (with respect to Banach measure in $N$) implies that Collatz conjecture fails. Let $\gamma_t(G)$ be the total domination number of graph $G$, a graph $G$ is $k$-total domination vertex critical (or\ just\ $k$-$\gamma_t$-critical) if $\gamma_t(G)=k$, and for any vertex $v$ of $G$ that is not adjacent to a vertex of degree one, $\gamma_t(G-v)=k-1$. Mojdeh and Rad \cite{MR06} proposed an open problem: Does there exist a 3-$\gamma_t$-critical graph $G$ of order $\Delta(G)+3$ with $\Delta(G)$ odd? Intracellular levels of glycerol-3-phosphate dehydrogenase (G-3-PDH) in baker's yeasts were monitored during storage at 3 different temperatures. Similar values for final biomass and specific activity of the enzyme were found, in each of two strains of baker's yeast, after 48 hours growth. The best medium tested, for the induction of G-3-PDH, contained: yeast extract (1% w/v), peptone (2% w/v), glycerol (3% w/v)and ethanol (1% w/v). Osmotic shock, provoked by suspending cells, after 48 hours growth in inducing medium, in 0.6 M NaCl solution for 2 hours, caused the activity to increase by a factor of 1.2. Cells of the GDO yeast strain, grown in inducing conditions, washed and lyophilized, exhibited a 35% rise in G-3-PDH activity during storage (10 days) at ambient temperature (about 27 ºC). Both refrigeration (4-5 ºC) and freezer storage (-18 ºC) maintained the G-3-PDH activity for up to 8 months.; Níveis intracelulares de G-3-PDH (sn-glicerol-3-fosfato:NAD+ 2oxidoredutase, EC 1.1.1.8) de levedura de panificação foram acompanhados durante a estocagem sob três diferentes temperaturas. El propósito de este trabajo es evaluar los efectos del Acuerdo Comercial "El Grupo de los Tres" (G-3) sobre el mercado automotriz colombiano. Asumiendo que la demanda se puede describir como un modelo de elección discreta, se desarrolla un modelo de equilibrio parcial para analizar la cuota de importación impuesta a los vehículos provenientes de México y su efecto sobre los precios en el mercado de automóviles en Colombia. Se encuentra que aunque la cuota redujo el promedio de los precios del mercado, este efecto no fue mayor a un 3% con respecto al escenario sin G-3. Asimismo, la cuota redujo únicamente los precios de los vehículos mexicanos, los cuales cayeron un 20%. Finalmente, se encuentra evidencia de que la distribución de la reducción de precios entre los segmentos es independiente de las participaciones del mercado. Por lo tanto, la cuota no muestra ser la mejor herramienta de política para cumplir los intereses domésticos. Estos resultados son relevantes para el diseño de políticas comerciales en industrias específicas y para la toma estratégica de decisiones en las firmas pertenecientes a la industria automotriz.

Introducción y Objetivos: Teniendo en cuenta la importancia sanitaria del aporte de ácidos grasos omega-3 y omega-6, y ante la escasez de estudios sobre el tema en colectivos españoles, se plantea la conveniencia de conocer la ingesta de estos ácidos grasos, su adecuación a los objetivos nutricionales marcados y sus fuentes alimentarias en una muestra representativa de la población española. Métodos: Se ha estudiado un colectivo de 1068 adultos (521 varones y 547 mujeres) de 17 a 60 años, seleccionados en diez provincias españolas, que constituyen una muestra representativa de la población, a nivel nacional. Se determino la ingesta de ácidos grasos poliinsaturados (AGP), ácidos grasos omega-3, α-linolénico (ALA), ácido eicosapentaenoico (EPA) y docosahexaenoico (DHA), ácidos grasos omega-6, ácido linoléico (LA) y araquidónico, en g/día y en porcentaje de la energía, utilizando un "Registro del consumo de alimentos" durante 3 días consecutivos, incluyendo un domingo, y recogiendo también datos personales, sanitarios y antropométricos de los individuos estudiados.
