Herein we report an approach to the formation of 5-alkynyl-1,3-dioxin-4-ones using Suzuki-Miyaura cross-coupling reaction of potassium alkynyltrifluoroborate salts with 2,2,6-trimethy1-5-iodo-1,3-dioxin-4-one. The resulting 5-ethynyltrimethylsilyl-1,3-dioxin-4-ones obtained through the Sonogashira reaction were further reacted in a Cu(I)-catalyzed Huisgen azide-alkyne 1,3-dipolar cycloaddition to form functionalized 1,4-disubstituted-1,2,3-triazoles in good yields, using mild conditions and ultrasonic radiation to expedite the reaction. An optimization study of the reaction conditions of Fe(TDCPP)Cl when it is used as catalyst in the hydroxylation of cyclohexane by iodosylbenzene (PhIO) has been carried out, It was found that Fe(TDCPP)Cl follows the classical PhIO mechanism described for Fe(TPP)Cl, which involves the monomeric active species Fe-IV(O)P-+. (I). In the optimized condition ([Fe(TDCPP) = 3.0 X 10(-4) mol l(-1) in 1,2-dichloroethane (DCE); ultrasound stirring at 0 degrees C; PhIO/FeP molar ratio = 100), this FeP led to a yield of cyclohexanol (C-ol) of 96% and a turnover number of 96, Therefore, Fe(TDCPP)Cl may be considered a good biomimetic model and a very stable, resistant and selective catalyst, which yields C-ol as the sole product. DCE showed to be a better solvent than dichloromethane (DCM), 1 DCE:1 MeOH mixture or acetonitrile (ACN). Since the Fe-IV(O)P-+. is capable of abstracting hydrogen atom from DCM, MeOH or ACN, the solvent competes with the substrate. Anti-inflammatory drugs are known to be the most widely-marketed drugs in the world, despite their serious side effects, mainly on the gastrointestinal tract. Thus, there are constant efforts to discover new prototypes with improved therapeutic activity and safety for the patient. Since the advent of the computational chemistry, the theoretical study of the physiological behavior of a new compound and hence an understanding of its supposed mechanism of action have been made a lot more accessible. The title compound, C16H36N+ C7H5O4 ÿ C7H6O4 or (nBu4N)+ (2,6-dhb) ÿ (2,6±Hdhb) [where (nBu4N) is tetrabutylammonium, (2,6-dhb) ÿ is 2,6-dihydroxybenzoate and (2,6-Hdhb) is 2,6-dihydroxybenzoic acid], crystallizes in the space group P1. The crystal structure involves alternate layers, in the c direction, of cationic [nBu4N]n n+ and anionic [(2,6- dhb)(2,6-Hdhb)]n nÿ . Hydrogen bonds within the anionic layers form bridges between 2,6-Hdhb and 2,6-dhb ÿ residues.

Although a large number of crystal structures of heterocyclic thiosemicarbazones have recently appeared in the literature, few structures of heterocyclic bis(thiosemicarbazones) or their metal complexes have been reported. Complexes of iron(II), indium(III), tin(IV), bismuth(III) involve bis(thiosemicarbazones) coordinating as N3S2 pentadentate ligands, often resulting in 7-coordinate complexes. In contrast, complexes with zinc(II) are often binuclear with thiosemicarbazone moieties of a bis(thiosemicarbazone) coordinating to two different zinc centers. Also included in this study is the structure of the first complex with a 2,6-diformylpyridine bis(thiosemicarbazone) ligand, a 4-coordinate nickel(II) complex with unusual coordination.

In this work, a new chiral stationary phase heptakis (3-O-pentafluoropropionyl-2,6-di-O-pentyl)-β-cyclodextrin was synthesized and evaluated with a wide variety of volatile chiral compounds. As a result, the new cyclodextrin derivative can be applied to the separation of chiral components of essential oils, as hydrocarbons, ketones, aldehydes, alcohols and of various classes of synthetic chiral compounds, as long chain alcohols, halocarbons, nitrogen and sulfur containing compounds.

6-Phosphofructo-2-kinase (EC 2.7.1.105)/fructose-2,6-bis-phosphatase (EC 3.1.3.46) catalyzes the synthesis and degradation of fructose 2,6-bisphosphate, a ubiquitous stimulator of glycolysis. The liver (L-type) and muscle (M-type) mRNAs for this bifunctional enzyme arise from distinct promoters of the same gene. We have now characterized in rat hepatoma FTO2B cells another mRNA, which is transcribed from a third promoter of that gene. This F-type mRNA is present in fetal rat liver and muscle, in rat placenta, and in several established rat cell lines. The F promoter contains no TATA box but contains several binding sites for Sp1 and for members of the ets oncogene family. Transfection of FTO2B cells with constructs containing the intact or mutagenized F promoter showed that its activity depends mainly on one of these sites. This site bound a heteromeric FTO2B cell protein indistinguishable from the ets-related GA binding protein alpha/ankyrin-repeats GA binding protein beta transcription factor.

Epstein-Barr virus nuclear antigens (EBNAs) are expressed in a cell-phenotype-dependent manner. EBNA 1 is regularly expressed in all Epstein-Barr virus-carrying cells, whereas EBNAs 2-6 are only expressed in Epstein-Barr virus-carrying cells with a lymphoblastoid phenotype including group III Burkitt lymphoma (BL) lines positive for B-cell activation markers. Transcripts are initiated at the BCR2 or exceptionally at one BWR1 promoter in lymphoblastoid cell lines and group III BL lines. In group I BL lines, nasopharyngeal carcinoma, and the somatic cell hybrids, where EBNAs 2-6 are downregulated, the BCR2/BWR1 promoter complex is inactive or switched off. Upregulation of EBNAs 2-6 in group III BL cells and in 5-azacytidine-treated group I BL cells accompanies the activation of the silent BCR2/BWR1 promoters. Activation of BCR2 parallels demethylation of at least one CpG pair in the same promoter region. The activity of BCR2/BWR1 promoter complex depends on a particular B-cell phenotype. EBNA 1 transcription must be initiated at another promoter in cells that express only EBNA 1.

31P-NMR spectroscopy was used to identify reaction intermediates during catalytic turn-over of the fructose-2,6-bisphosphatase domain (Fru-2,6-P2ase) of the bifunctional enzyme, 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase. When fructose-2,6-bisphosphate (Fru-2,6-P2) was added to the enzyme, the 31P-NMR spectrum showed three resonances in addition to those of free substrate: the phosphohistidine (His-P) intermediate, the C-6 phosphoryl group of fructose-6-phosphate bound to the phosphoenzyme, and phosphate generated by the hydrolysis of substrate. Direct analysis of the alkali-denatured phospho-enzyme intermediate by 1H-31P heteronuclear multiple quantum-filtered coherence spectroscopy confirmed the formation of 3-N-phosphohistidine. Binding of fructose 6-phosphate to the bisphosphatase was detected by a down-field shift and broadening of the C-6 phosphoryl resonance. The down-field shift was greater in the presence of the phosphoenzyme intermediate. Inhibition of Fru-2,6-P2 hydrolysis by fructose 6-phosphate and Fru-2,6-P2 was shown to involve binding of the sugar phosphates to the phosphoenzyme. This study provides new experimental evidence in support of the reaction mechanism of Fru-2,6-P2ase and suggests that the steady-state His-P intermediate exists primarily in the E-P.fructose 6-phosphate complex. These results lay a solid foundation for the use of 31P-NMR magnetization transfer studies to provide an in-depth analysis of the bisphosphatase reaction mechanism.

Liver and skeletal muscle isoforms of 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase (6PF2K/Fru-2,6-P(2)ase) isoenzymes are products of alternatively spliced first exons of the same gene, with common kinase and bisphosphatase domains. The muscle-specific exon-1 encodes nine unique amino acids, that lack the cAMP-dependent protein kinase (PK-A) phosphorylation site, and differ in sequence from those encoded by the liver-specific exon-1 (32 amino acids), contributing to its much lower affinity for fructose 6-phosphate (Fru-6-P). PK-A phosphorylation of the liver isoform at Ser(32) reduces the affinity of the kinase for Fru-6-P, and stimulates the bisphosphatase V(max). In the present study, we have defined the locus of interaction of the N-terminal residues with the N-terminal kinase and C-terminal domains by successive N- and C-terminal deletions. This study shows that: (1) residues Gly(5)-Glu(6)-Leu(7) of the liver isoform are responsible for increasing the affinity of 6PF2K for Fru-6-P, maintaining the inhibition of Fru-2,6-P(2)ase activity, and mediating the effects of PK-A phosphorylation on the two activities; (2) the loss of Fru-6-P inhibition of the bisphosphatase and the enhancement of its V(max), rather than the inhibition of the kinase...

The hepatic bifunctional enzyme, 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase (6PF-2-K/Fru-2,6-P2ase), E.C. 2.7-1-105/E.C. 3-1-3-46, is one member of a family of unique bifunctional proteins that catalyze the synthesis and degradation of the regulatory metabolite fructose-2,6-bisphosphate (Fru-2,6-P2). Fru-2,6-P2 is a potent activator of the glycolytic enzyme 6-phosphofructo-1-kinase and an inhibitor of the gluconeogenic enzyme fructose-1,6-bisphosphatase, and provides a switching mechanism between these two opposing pathways of hepatic carbohydrate metabolism. The activities of the hepatic 6PF-2-K/Fru-2,6-P2ase isoform are reciprocally regulated by a cyclic AMP-dependent protein kinase (cAPK)-catalyzed phosphorylation at a single NH2-terminal residue, Ser-32. Phosphorylation at Ser-32 inhibits the kinase and activates the bisphosphatase, in part through an electrostatic mechanism. Substitution of Asp for Ser-32 mimics the effects of cAPK-catalyzed phosphorylation. Aberrant cell-surface glycosylation patterns are present on virtually all tumors and have been linked to tumor progression, metastasis, and invasivity. We have shown that expressing a normally quiescent, glycoprotein-specific α2,6-sialyltransferase (ST6Gal1) gene in gliomas inhibited invasivity in vitro and tumor formation in vivo. To identify other glycogene targets with therapeutic potential, we created a focused 45-mer oligonucleotide microarray platform representing all of the cloned human glycotranscriptome and examined the glycogene expression profiles of 10 normal human brain specimens, 10 malignant gliomas, and 7 human glioma cell lines. Among the many significant changes in glycogene expression observed, of particular interest was the observation that an additional α2,6-sialyltransferase, ST6 (α-N-acetyl-neuraminyl-2,3-β-galactosyl-1,3)-N-acetylgalactosaminide α2,6-sialyltransferase 5 (ST6GalNAcV), was expressed at very low levels in all glioma and glioma cell lines examined compared with normal brain. The title complex, [CuCl(C12H8N2)2][Mn(C7H3NO4)Cl(C12H8N2)]·CH3OH, consists of discrete [CuCl(phen)2]+ cations (phen is 1,10-phenanthroline), [MnCl(pydc)(phen)]− anions (H2pydc is 2,6-pyridine-2,6-di­carb­oxy­lic acid) and one methanol solvent mol­ecule of crystallization per asymmetric unit. It should be noted, that a solvent-masking procedure as implemented in OLEX2 [Dolomanov et al. (2009). J. Appl. Cryst. 42, 339–341 ▶] was used to remove the electronic contribution from one disordered solvent molecule, presumably methanol. Only the atoms used in the refined model are reported in chemical formula and related values. The CuII ion is five-coordinated by two phenanthroline ligands and one chloride ion in a distorted trigonal–bipyramidal geometry. The dihedral angle between the phen ligands is 65.21 (5)°. During early postnatal development, the intestine is highly responsive to LR3IGF-I administration but refractory to IGF-I, in contrast to the mature intestine. Given that LR3IGF-I is an IGF-I analog that binds poorly to IGF binding proteins, the response of the intestine is likely to reflect regulation of IGF-I bioactivity by IGF binding proteins. This study measures the delivery of exogenous IGF-I peptides to the intestine in preweaning (d-19) and adult rats to determine whether a correlation exists with the potency advantage of LR3IGF-I in the intestine during postnatal development. IGF-I or LR3IGF-I (2.6 µg/kg) was spiked with corresponding 125I-labeled peptide (10 x 106 cpm) and administered iv as a bolus (n = 5–6/group) with blood and tissue samples collected 5 and 10 min post injection. In both age groups, the levels of 125I-IGF-I retained in the blood at both 5 and 10 min were higher than the levels of 125I-LR3IGF-I, consistent with the slower clearance rate for the native peptide. In the gastrointestinal tract, the levels of 125I-LR3IGF-I per gram of tissue were 37–50% higher than 125I-IGF-I. Surprisingly, there was little difference in the relative delivery of LR3IGF-I to IGF-I to the intestine, across developmental age. Partitioning and molecular dynamics of 2,2,6,6,-tetramethylpiperedine-1-oxyl (TEMPO) nitroxide radicals in large unilamellar liposomes (LUV) composed from 1,2-dipalmitoyl-sn-glycero-3-phosphatidylcholine were investigated by using very high frequency electron paramagnetic resonance (EPR) spectroscopy. Experiments carried out at a microwave frequency of 94.3 GHz completely resolved the TEMPO EPR spectrum in the aqueous and hydrocarbon phases. An accurate computer simulation method combined with Levenberg-Marquardt optimization was used to analyze the TEMPO EPR spectra in both phases. Spectral parameters extracted from the simulations gave the actual partitioning of the TEMPO probe between the LUV hydrocarbon and aqueous phases and allowed analysis of picosecond rotational dynamics of the probe in the LUV hydrocarbon phase. In very high frequency EPR experiments, phase transitions in the LUV-TEMPO system were observed as sharp changes in both partitioning and rotational correlation times of the TEMPO probe. The phase transition temperatures (40.5 +/- 0.2 and 32.7 +/- 0.5 degrees C) are in agreement with previously reported differential scanning microcalorimetry data. Consultable des del TDX; Descripció del recurs: 10 de febrer 2005; Els treballs que inclou aquest compendi s'han desenvolupat dins del projecte FEDER 2FD97-0916-C02-02, que té com a objectiu principal confirmar l'existència, en una població comercial Landrace altament seleccionada, de quantitative trait loci (QTL) prèviament descrits en encreuaments de races divergents. Els caràcters analitzats en aquest projecte inclouen tant mesures de la qualitat de la carn com de conformació i de qualitat de la canal. Mitjançant la segregació de marcadors moleculars tipus microsatèl·lits en un pedigrí de dues generacions d'una població Landrace es van analitzar 10 regions dels cromosomes 1, 2, 3, 4, 6, 7, 8, 9, 10 i 13. Els resultats demostren l'existència d'efectes als cromosomes 2, 6, 7, 8, 9 i 10, tot indicant que QTL descrits prèviament en encreuaments de races divergents són encara presents en les poblacions comercials. A més a més s'han caracteritzat quatre gens candidats (MDH1, ME1, DECR i ACLS1) relacionats amb el metabolisme lipídic. Encara que en tots quatre s'han pogut identificar polimorfismes, només en el cas del DECR les dues transversions detectades provoquen canvis aminoacídics. Els polimorfismes identificats en l'ME1 i el DECR s'han fet servir de marcadors en dos estudis d'associació. Descripció del recurs: 12 abril 2013; El conjunt d'articles que es presenten en aquesta tesi són el resultat d'un llarg període d'investigació científica sobre la Història de l'Empresa industrial i de la innovació tecnològica, centrada en la ciutat de Manresa. Les aportacions principals de la tesi giren al voltant de la història de l'empresa, considerada com l'agent econòmic en el que es prenen les decisions, la innovació tecnológica i té com a marc conceptual el districte industrial. La tesi està composta per 6 publicacions, 4 d'elles a la part fonamental i 2 a l'annex. En primer lloc, s'interessa pel funcionament d'una economia local, partint de la base de què Manresa i el seu hinterland han funcionat com un districte industrial en el sentit ampli del terme, que inclouria els districtes amb diversos sectors que funcionen alhora (el tèxtil i alguns dels seus subsectors especialitzats, al costat d'una metal·lúrgia originada com a indústria complementària del tèxtil i que acaba especialitzada en auxiliar de l'automòbil) i amb algunes empreses que funcionen com a hub. Avaluació geriàtrica en pacients nonagenaris: factors pronòstics, intervencions interdisciplinàries i eficàcia sobre la capacitat funcional Objectius: 1.Demostrar si els pacients nonagenaris milloren la seva capacitat funcional durant l'ingrés en una unitat de convalescència geriàtrica. 2.Identificar els factors pronòstics de guany funcional, de mortalitat i d'institucionalització. 3.Elaborar un instrument predictiu de guany funcional i fer una validació pilot del mateix instrument. Pacients i mètode: estudi prospectiu de 212 pacients nonagenaris (75,94% dones; edat mitjana 92,02 anys), que van ingressar en la unitat durant el període de 6 anys. A tots els pacients se'ls va fer una avaluació geriàtrica integral, un programa d'intervencions interdisciplinàries i quan va estar indicat un programa de rehabilitació (fisioteràpia i teràpia ocupacional). Variables registrades: edat, sexe, procedència, antecedents patològics, nombre de fàrmacs previs a l'ingrés i a l'alta, capacitat funcional prèvia (índex de Lawton i de Barthel), en el moment de l'ingrés i a l'alta (índex de Barthel), capacitat i característiques de la marxa prèvies i en el moment de l'alta, convivència i suport social previs, diagnòstic principal a l'ingrés...

Descripció del recurs: el 7 de gener de 2011; L'objectiu d'aquesta tesi ha estat avaluar diferents dispositius de identificació del bestiar cabrum, recentment contemplades a la legislació europea. S'han portat a terme 4 experiments per tal d'avaluar els següents dispositius: cròtals plàstics i de ràdio freqüència (RFID), bols ruminals, i identificadors a les extremitats (injectables i braçalets). A l'Exp. 1 es van emprar 97 cabrits de raça Murciano-Granadina per estudiar l'aplicació i capacitat de lectura (CL) de cròtals visuals (V1: tip-tag; V2: oficial), cròtals de RFID (E1: bandera-botó; E2: botó), mini-bols (B1: 13,8 g; B2: 20 g), i injectables a l'extremitat anterior (T1: 15 mm; T2: 12 mm); les mares (n = 29) es van identificar amb bols de mida estàndard (B3: 75 g). Al cap d'un any, la CL de B3, E1 i E2 va ser del 100%. Les CL més baixes es van donar amb B1 (71,4%) i V1 (82,9%). En conclusió, només els cròtals de RFID tipus botó van permetre la adequada ID de cabrides de reposició de raça lletera. A l'Exp. 2, es van emprar 295 cabres adultes i segalles de races Alpina, Angora, Boer i Spanish, explotades en condicions semi-extensives.
