L-NAME-treatment alters ectonucleotidase activities in kidney membranes of rats

Aims: To investigate the effect of N omega-Nitro-L-arginine methyl ester CL-NAME) treatment, known to induce a sustained elevation of blood pressure, on ectonucleotidase activities in kidney membranes of rats. Main methods: L-NAME (30 mg/kg/day) was administered to Wistar rats for 14 days in the drinking water. Enzyme activities were determined colorimetrically and their gene expression patterns were analyzed by semi-quantitative RT-PCR. The metabolism of ATP and the accumulation of adenosine were evaluated by HPLC in kidney membranes from control and hypertensive rats. PKC phosphorylation state was investigated by Western blot. Key findings: We observed an increase in systolic blood pressure from 115 +/- 12 mmHg (control group) to 152 18 mmHg (L-NAME-treated group). Furthermore, the hydrolysis of ATP, ADP, AMP, and p-Nph-5`TMP was also increased (17%, 35%, 27%, 20%, respectively) as was the gene expression of NTPDase2, NTPDase3 and NPP3 in kidneys of hypertensive animals. Phospho-PKC was increased in hypertensive rats. Significance: The general increase in ATP hydrolysis and in ecto-5`-nucleotidase activity suggests a rise in renal adenosine levels and in renal autoregulatory responses in order to protect the kidney against the threat presented by hypertension. It is well known that hypertension is closely associated to the development of vascular diseases and that the inhibition of nitric oxide biosynthesis by administration of N omega-Nitro-L-arginine methyl ester hydrochloride (L-NAME) leads to arterial hypertension. In the vascular system, extracellular purines mediate several effects: thus, ADP is the most important platelet agonist and recruiting agent, while adenosine, all end product Of nucleotide metabolism, is a vasodilator and inhibitor of platelet activation and recruitment. Members of several families of enzymes, known as ectonucleotidases, including E-NTPDases (ecto-nucleoside triphosphate diphosphohydrolase), E-NPP (ecto-nucleotide pyrophosphatase/phosphodiesterase) and 5`-nucleotidase are able to hydrolyze extracellular nucleotides until their respective nucleosides. Entretanto, por coletar dados de diferentes fontes, surge o problema de informações ambíguas ou duplicadas de nomes de autores. Métodos tradicionais de desambiguação de nomes utilizam informação sintática de atributos. Todavia, recentemente o uso de redes de relacionamentos, que traz informação semântica, tem sido estudado em desambiguação de dados. Em desambiguação de nomes de autores, relações de co-autoria podem ser usadas para criar uma rede social, que pode ser utilizada para melhorar métodos de desambiguação de nomes de autores. This study determined the protective effect of silibinin, a flavonoid with anti-inflammatory and hepatoprotective properties on the deleterious effects observed in experimentally induced preeclampsia in rats.Main methods: Pregnant Wistar rats were treated during gestation (days 10-20) with L-NAME (70-80 mg/kg/day) in drinking water or with L-NAME plus silibinin (100 mg/kg/day, orally) starting at day 0, day 7 or day 14 of pregnancy. Male Holtzman rats weighting 200-250 g were anesthetized with zoletil 50 mg/Kg (tiletamine chloridrate 125,0 mg and zolazepan chloridrate 125,0 mg) into quadriceps muscle and submitted an electrolytic lesion of the lateral hypothalamus (LH) and a stainless steel cannula was implanted into their median preoptic nucleus (MnPO). We investigated the effects of the injection into the (MnPO) of FK 409 (20 mug/0.5 mul), a nitric oxide (NO) donor, and N-W-nitro-L-arginine methyl ester (L-NAME) 40 mug/0.5 mul, a nitric oxide synthase inhibitor (NOSI), on the water and sodium appetite and the natriuretic, diuretic and cardiovascular effects induced by injection of L-NAME and FK 409 injected into MnPO in rats with LH lesions. Controls were injected with a similar volume of 0.15 M NaCl. L-NAME injected into MnPO produced an increase in water and sodium intake and in sodium and urine excretion and increase de mean arterial pressure (MAP). Paw edema was induced in male Wistar rats (200-250 g) by intraplantar (ipl) administration of 2.5 mu g endotoxin (Etx). Etx, like carrageenin, produced two distinct edema formation phases, an early phase (75 min) followed by a late phase (7 h). We showed that the edema formation in the early phase was antagonized by dipyrone (80 mg/kg, ip) and indomethacin (1 mg/kg, ip) by 52% and 55%, respectively, and that the late phase was resistant to these drugs. These results suggest that in the early phase prostaglandins appear to be involved in the process. However, the activation of the kinin cascade leading to the release of other mediators may be involved in the increase of edema in the late phase. To test this hypothesis, we investigated whether the release of nitric oxide (NO) is involved in the mechanism of endotoxin-induced rat paw edema during the late phase, using N omega-nitro-L-arginine methyl ester (L-NAME) (50 mu g, ipl) as inhibitor of NO synthase and L-arginine (1 mg, ipl) as substrate of NO synthase. The paw edema induced by Etx was inhibited by L-NAME by 56% and increased by L-arginine by 81%. Furthermore, L-arginine given in combination with L-NAME completely reversed the inhibition of Etx-induced edema produced by L-NAME. These results support the hypothesis that in the late phase NO production is associated with the edema evoked by Etx.

O óxido nítrico (NO) é uma molécula gasosa, de vida curta, que modula a neurotransmissão, a força de contração, o metabolismo de glicose, o tônus vascular e a função muscular em geral. O músculo esquelético é a maior fonte de NO em mamíferos e expressa todas as três isoformas de NO sintases (NOS). Os inibidores da NOS são ferramentas usadas para estudar a função do NO na fisiologia muscular. Neste trabalho, nós examinamos os efeitos do tratamento crônico de ratos com N?-nitro-L-arginina metil éster (L-NAME), que bloqueia a síntese de NO pelas NOSs, através de parâmetros bioquímicos e morfológicos dos músculos soleus e extensor digitorum longus. A tipagem das fibras musculares, a medida da área de secção transversal das fibras, a expressão das isoformas de miosina, a determinação do número dos núcleos da fibra muscular e, a expressão e imunolocalização da NOS endotelial e neuronal foram parâmetros usados para avaliar os efeitos da falta de NO nos músculos. Os tendões são estruturas que transmitem a força muscular ao osso. São formados por células imersas em uma matriz extracelular rica em fibras de colágeno paralelas. Tendões, como o calcanear, são resistentes, contudo podem ser acometidos por lesões. Esse tecido lesado não se regenera totalmente e a organização, estrutura e propriedades mecânicas do tendão reparado são inferiores ao tendão saudável. Além disso, há formação de adesão entre o tendão e o tecido conjuntivo adjacente, o que constitui um problema clínico. Tem sido reportado que o oxido nítrico (NO) tem influência na regeneração do tecido e que a inibição da síntese de NO poderia estar diretamente relacionada a uma não recuperação do tecido lesado. Dessa forma, este trabalho teve como objetivo analisar o dano estrutural e o acúmulo de células inflamatórias após a injúria em tendão calcanear de ratos adultos não tratados e tratados com o inibidor da oxido nítrico sintetase (NOS), L-NAME (Nω-nitro-L-arginina metil éster). The aim of the present study was to investigate the role of bradykinin in the inhibitory action of captopril in hypertension induced by L-NAME in anesthetized rats. Male Wistar rats (260-320 g) were anesthetized with chloralose and arterial blood pressure was recorded with a polygraph pressure transducer. The hypertensive effect of L-NAME was studied in rats pretreated with saline, captopril or HOE 140 plus captopril. The effect of captopril was also studied during the sustained pressor effect of L-NAME. The acute pressor effect of L-NAME (10 mg/kg, iv) was significantly reduced by iv pretreatment with 2 mg/kg captopril (D increase of 49 ± 4.9 mmHg reduced to 20 ± 5.4 mmHg, P = 0.01). The pressor effect of L-NAME (D increase of 38 ± 4.8 mmHg) observed in rats pretreated with captopril and HOE 140 (0.1 mg/kg, iv) was not significantly different from that induced by L-NAME in rats pretreated with saline (P = 0.09). During the sustained pressor effect induced by L-NAME (D increase of 49 ± 4.9 mmHg) captopril induced a significant (P<0.05) reduction in arterial blood pressure (D decrease of 22 ± 3.0 mmHg). The present results demonstrate that the acute pressor effect of L-NAME is reduced by captopril and this inhibitory effect may be partly dependent on the potentiation of the vasodilator actions of bradykinin

We evaluated the hemodynamic pattern and the contribution of the sympathetic nervous system in conscious and anesthetized (1.4 g/kg urethane, iv) Wistar rats with L-NAME-induced hypertension (20 mg/kg daily). The basal hemodynamic profile was similar for hypertensive animals, conscious (N = 12) or anesthetized (N = 12) treated with L-NAME for 2 or 7 days: increase of total peripheral resistance associated with a decrease of cardiac output (CO) compared to normotensive animals, conscious (N = 14) or anesthetized (N = 14). Sympathetic blockade with hexamethonium essentially caused a decrease in total peripheral resistance in hypertensive animals (conscious, 2 days: from (means ± SEM) 2.47 ± 0.08 to 2.14 ± 0.07; conscious, 7 days: from 2.85 ± 0.13 to 2.07 ± 0.33; anesthetized, 2 days: from 3.00 ± 0.09 to 1.83 ± 0.25 and anesthetized, 7 days: from 3.56 ± 0.11 to 1.53 ± 0.10 mmHg mL-1 min-1) with no change in CO in either group. However, in the normotensive group a fall in CO (conscious: from 125 ± 4.5 to 96 ± 4; anesthetized: from 118 ± 1.5 to 104 ± 5.5 mL/min) was observed. The responses after hexamethonium were more prominent in the hypertensive anesthetized group. However, no difference was observed between conscious and anesthetized normotensive rats in response to sympathetic blockade. The present study shows that the vasoconstriction in response to L-NAME was mediated by the sympathetic drive. The sympathetic tone plays an important role in the initiation and maintenance of hypertension.

In Brazil, HIV-infected individuals receive drugs (including non-brand name drugs which comprise locally produced generics and drugs that have not been tested in bioequivalence trials) free of charge from the government. The objective of the present study was to evaluate the effectiveness of highly active antiretroviral therapy (HAART) in Rio de Janeiro, Brazil, where non-brand drugs are widely used. For this purpose, we estimated the proportion of subjects with virologic failure (plasma HIV viral load greater than 400 copies/mL at 6 months after initiation of treatment). This was a retrospective cohort study of drug-naive HIV-infected subjects who initiated HAART. Subjects were included in the analysis if they were 18 years of age or older, were treatment naive, started HAART with a minimum of 3 drugs, and had available information on blood plasma HIV-1 viral load after 6 months on therapy. All subjects used antiretrovirals in dosing regimens recommended by the Brazilian National Advisory Committee for Antiretroviral Therapy. INTRODUCTION: The role of ovarian hormones and nitric oxide in learning and memory has been widely investigated. OBJECTIVE: The present study was carried out to evaluate the effect of the nitric oxide synthase (NOS) inhibitor, N (G)-nitro-L-arginine methyl ester (L-NAME), on the ability of estradiol to improve learning in OVX rats using the Morris water maze. METHODS: Forty rats were divided into five groups: (1) ovariectomized (OVX), (2) ovariectomized-estradiol (OVX-Est), (3) ovariectomized-L-NAME 10 (OVX-LN 10), (4) ovariectomized-L-NAME 50 (OVX-LN 50) and (5) ovariectomized-estradiol-L-NAME 50 (OVX-Est-LN 50). The animals in the OVX-Est group were treated with a weekly injection of estradiol valerate (2 mg/kg; i.m.). The OVX-LN 10 and OVX-LN 50 groups were treated with daily injections of 10 and 50 mg/kg L-NAME (i.p.), respectively. The animals in the OVX-Est-LN 50 group received a weekly injection of estradiol valerate and a daily injection of 50 mg/kg L-NAME. After 8 weeks, all animals were tested in the Morris water maze. RESULTS: The animals in the OVX-Est group had a significantly lower latency in the maze than the OVX group (p<0.001). There was no significant difference in latency between the OVX-LN 10 and OVX-LN 50 groups in comparison with the OVX group. This thesis examines the impact of a corporate name change on stock price and trading volume of Canadian companies around the announcement date, the approval date, and the adoption date over the time period from 1997 to 2011. Name changes are classified into six categories: major and minor, structural and pure, diversified and focused, accompanied with a change in ticker symbol and without a change in ticker symbol, “Gold” name addition and deletion, and different reasons for name changes (e.g., merger and acquisition, change of structure, change of strategy, and better image). The thesis uses the standard event study methodology to perform abnormal return and trading volume analyses. In addition, regression analysis is employed to examine which type of a name change has the largest impact on cumulative abnormal returns. Sample stocks exhibit a significant positive abnormal return one-day prior to the approval day and one day after the adoption date. Around the approval date we observe significant abnormal returns for stocks with a structural name change. The perceived simplicity of interprocess cooperation is strongly influenced by the type of name space in which the processes involved execute. If multiple processes share a common name space, they are more likely to cooperate effectively than if each operates in a distinct name space. This paper argues that, especially where specialized resources, such as a supercomputer, are involved, per-process name spaces are the optimum way to provide common name spaces across arbitrary domain boundaries. This paper also argues that such name spaces are best implemented in user mode. This paper analyzes the worst-case efficiency ratio of false-name-proof combinatorial auction mechanisms. False-name-proofness generalizes strategy-proofness by assuming that a bidder can submit multiple bids under fictitious identifiers. Even the well-known Vickrey-Clarke-Groves mechanism is not false-name-proof. It has previously been shown that there is no false-name-proof mechanism that always achieves a Pareto efficient allocation. Consequently, if false-name bids are possible, we need to sacrifice efficiency to some extent. This leaves the natural question of how much surplus must be sacrificed. To answer this question, this paper focuses on worst-case analysis. Specifically, we consider the fraction of the Pareto efficient surplus that we obtain and try to maximize this fraction in the worst-case, under the constraint of false-name-proofness. As far as we are aware, this is the first attempt to examine the worst-case efficiency of false-name-proof mechanisms. We show that the worst-case efficiency ratio of any false-name-proof mechanism that satisfies some apparently minor assumptions is at most 2/(m + 1) for auctions with m different goods. Übergewicht und Bluthochdruck stellen wichtige Risikofaktoren für die Entstehung kardiovaskulärer Erkrankungen dar. Das Hormon Leptin, welches von Fettzellen produziert wird und somit bei Adipositas im Serum erhöht ist, könnte möglicherweise ein Verbindungsglied zwischen arterieller Hypertonie und Adipositas darstellen. Ziel dieser Studie war es, die Rolle von NO auf Blutdruck und renale Hämodynamik bei übergewichtigen Probanden im Vergleich zu Normalpersonen zu untersuchen. 10 übergewichtige, normotensive, gesunde Männer (BMI>30 kg/m2) nahmen an der Studie teil und wurden mit 11 Normalgewichtigen (BMI<25 kg/m2), die durch eine Vorstudie mit gleichem Versuchsaufbau geliefert wurden, verglichen. Sie erhielten doppelblind intravenös den NO-Syntheseantagonisten L-NAME (2mg/kg Körpergewicht über 30 Minuten) oder Placebo. Objectives: The study sought to investigate how Spanish names are handled by national and international databases and to identify mistakes that can undermine the usefulness of these databases for locating and retrieving works by Spanish authors. Methods: The authors sampled 172 articles published by authors from the University of Granada Medical School between 1987 and 1996 and analyzed the variations in how each of their names was indexed in Science Citation Index (SCI), MEDLINE, and indice M6dico Espanol (IME). The number and types of variants that appeared for each author's name were recorded and compared across databases to identify inconsistencies in indexing practices. We analyzed the relationship between variability (number of variants of an author's name) and productivity (number of items the name was associated with as an author), the consequences for retrieval of information, and the most frequent indexing structures used for Spanish names. Results: The proportion of authors who appeared under more then one name was 48.1% in SCI, 50.7% in MEDLINE, and 69.0% in IME. INTRODUCTION: The role of ovarian hormones and nitric oxide in learning and memory has been widely investigated. OBJECTIVE: The present study was carried out to evaluate the effect of the nitric oxide synthase (NOS) inhibitor, N (G)-nitro-L-arginine methyl ester (L-NAME), on the ability of estradiol to improve learning in OVX rats using the Morris water maze. METHODS: Forty rats were divided into five groups: (1) ovariectomized (OVX), (2) ovariectomized-estradiol (OVX-Est), (3) ovariectomized-L-NAME 10 (OVX-LN 10), (4) ovariectomized-L-NAME 50 (OVX-LN 50) and (5) ovariectomized-estradiol-L-NAME 50 (OVX-Est-LN 50). The animals in the OVX-Est group were treated with a weekly injection of estradiol valerate (2 mg/kg; i.m.). The OVX-LN 10 and OVX-LN 50 groups were treated with daily injections of 10 and 50 mg/kg L-NAME (i.p.), respectively. The animals in the OVX-Est-LN 50 group received a weekly injection of estradiol valerate and a daily injection of 50 mg/kg L-NAME. After 8 weeks, all animals were tested in the Morris water maze. El ñame (Dioscorea alata L.), ampliamente usado como fuente de calorías en África, Asia y el Caribe, es una planta herbácea y trepadora que se reproduce generalmente de forma vegetativa y produce un tubérculo comestible de gran tamaño con alto contenido de almidón. Varios microorganismos fitopatógenos, incluyendo virus, hongos y bacterias, afectan al cultivo. Plantas enfermas de la zona de Guarataro, estado Bolívar, Venezuela, fueron colectadas en el 2003, año en que hubo ~80% de pérdidas en la producción de ñame de la zona a causa de una enfermedad desconocida. Hojas colectadas de plantas enfermas con lesiones acuosas y necróticas fueron esterilizadas superficialmente y transferidas a solución salina para permitir la difusión de bacterias desde los bordes del tejido y ser luego sembradas en medio LB agar. Como controles se emplearon agua y medio LB. Colonias amarillas, brillantes y lisas, fueron visibles a las 24-48h de incubación a 30ºC. Varios aislados fueron inoculados en las pruebas de hipersensibilidad en tabaco y de patogenicidad en plántulas de ñame cultivadas in vitro.
