Objetivo: métodos antropométricos que quantifiquem as curvas da coluna vertebral e a avaliação postural a fim de realizar investigações epidemiológicas sobre o papel da postura na ocorrência das dores lombares. O propósito do estudo foi avaliar acurácia e reprodutibilidade do Sistema de Avaliação Postural Digitalizado (SAPD) para medir lordose lombar comparando com raio-x. Delineamento: transversal, com amostra consecutiva. Participantes: para medida da acurácia no grupo 1 ( T12,L3,L5) n = 16 e grupo 2 (L1,L3,L5) n= 17. Na reprodutibilidade intra e inter-avaliador n= 80. Principais Medidas: marcadores externos nos processos espinhosos das vértebras T12, L1, L3 e L5. Raio-x de perfil da coluna lombar e foto digital em perfil direito. Medida da lordose lombar no raio-x com métodos de Cobb,Centróide (CLL) e Processos Espinhosos (PE) e com o SAPD. Campylobacter fetus bacteria, isolated from both mammals and reptiles, may be either subsp. fetus or subsp. venerealis and either serotype A or serotype B. Surface layer proteins, expressed and secreted by genes in the sap locus, play an important role in C. fetus virulence. To assess whether the sap locus represents a pathogenicity island and to gain further insights into C. fetus evolution, we examined several C. fetus genes in 18 isolates. All of the isolates had 5 to 9 sapA or sapB homologs. One strain (85-387) possessed both sapA and sapB homologs, suggesting a recombinational event in the sap locus between sapA and sapB strains. When we amplified and analyzed nucleotide sequences from portions of housekeeping gene recA (501 bp) and sapD (450 bp), a part of the 6-kb sap invertible element, the phylogenies of the genes were highly parallel. Among the 15 isolates from mammals, serotype A and serotype B strains generally had consistent positions. The fact that the serotype A C. fetus subsp. fetus and subsp. venerealis strains were on the same branch suggests that their differentiation occurred after the type A-type B split. Isolates from mammals and reptiles formed two distinct tight phylogenetic clusters that were well separated. Sequence analysis of 16S rRNA showed that the reptile strains form a distinct phylotype between mammalian C. fetus and Campylobacter hyointestinalis. The virulence of Campylobacter fetus, a bacterial pathogen of ungulates and humans, is mediated in part by the presence of a paracrystalline surface layer (S-layer) that confers serum resistance. The subunits of the S-layer are S-layer proteins (SLPs) that are secreted in the absence of an N-terminal signal sequence and attach to either type A or B C. fetus lipopolysaccharide in a serospecific manner. Antigenic variation of multiple SLPs (encoded by sapA homologs) of type A strain 23D occurs by inversion of a promoter-containing DNA element flanked by two sapA homologs. Cloning and sequencing of the entire 6.2-kb invertible region from C. fetus 23D revealed a probable 5.6-kb operon of four overlapping genes (sapCDEF, with sizes of 1,035, 1,752, 1,284, and 1,302 bp, respectively) transcribed in the opposite direction from sapA. The four genes also were present in the invertible region of type B strain 84-107 and were virtually identical to their counterparts in the type A strain. Pasteurella multocida was grown in iron-free chemically defined medium supplemented with hemoglobin, transferrin, ferritin, and ferric citrate as iron sources. Whole-genome DNA microarrays were used to monitor global gene expression over seven time points after the addition of the defined iron source to the medium. This resulted in a set of data containing over 338,000 gene expression observations. On average, 12% of P. multocida genes were differentially expressed under any single condition. A majority of these genes encoded P. multocida proteins that were involved in either transport and binding or were annotated as hypothetical proteins. Several trends are evident when the data from different iron sources are compared. In general, only two genes (ptsN and sapD) were expressed at elevated levels under all of the conditions tested. The results also show that genes with increased expression in the presence of hemoglobin did not respond to transferrin or ferritin as an iron source. Correspondingly, genes with increased expression in the transferrin and ferritin experiments were expressed at reduced levels when hemoglobin was supplied as the sole iron source. The fluorescent phorbol ester 12-N-methylanthraniloylphorbol 13-acetate [sapintoxin D (SAPD)] was used as both the activator and the probe for the activating conformational change of the C1 domain of recombinant protein kinase C (PKC)alpha. Fluorescence emission spectra and steady-state anisotropy measurements of SAPD in fully active membrane-associated PKC show that there is a relatively hydrophobic environment and restricted motional freedom characterizing the phorbol-ester-binding site. SAPD also interacts with the membrane lipids so that it was necessary to resort to time-resolved anisotropy measurements to resolve the signals corresponding to PKC-bound SAPD from that associated with buffer and lipid. In the presence of membrane lipids (unilamellar vesicles of phosphatidylcholine and phosphatidylserine, 4:1 molar ratio) and Ca(2+), at a concentration sufficient to activate the enzyme fully, a long correlation time characteristic of highly restricted motion was observed for PKC-associated SAPD. The fraction of SAPD molecules displaying this restricted motion, in comparison with the total SAPD including that in lipids and in buffer, increased with increasing concentrations of Ca(2+) and paralleled the appearance of enzyme activity...

CCAAT/enhancer binding proteinδ (C/EBPδ) plays a key role in mammary epithelial cell G0 growth arrest, and “loss of function” alterations in C/EBPδ have been reported in breast cancer and acute myeloid leukemia. C/EBPδ is regulated at the transcriptional, post-transcriptional, and post-translational levels, suggesting tight control of C/EBPδ content and function. Protein inhibitors of activated STATs (PIASs) regulate a growing number of transcription factors, including C/EBPs. HC11 nontransformed mammary epithelial cells express PIAS3, PIASxβ, and PIASy, and all three PIAS family members repress C/EBPδ transcriptional activity. PIASy is the most potent, however, repressing C/EBPδ transcriptional activity by >80%. PIASy repression of C/EBPδ transcriptional activity is dependent upon interaction between the highly conserved PIASy N-terminal nuclear matrix binding domain (SAPD) and the C/EBPδ transactivation domain (TAD). PIASy repression of C/EBPδ transcriptional activity is independent of histone deacetylase activity, PIASy E3 SUMO ligase activity, and C/EBPδ sumoylation status. PIASy expression is associated with C/EBPδ translocation from nuclear foci, where C/EBPδ co-localizes with p300, to the nuclear periphery. PIASy-mediated translocation of C/EBPδ is dependent upon the PIASy SAPD and C/EBPδ TAD. PIASy reduces the expression of C/EBPδ adhesion-related target genes and enhances repopulation of open areas within a cell monolayer in the in vitro “scratch” assay. Measurement of Disability is one of the off-shoot projects of the major multicentred study on ‘Factors Affecting Course and Outcome of Schizophrenia’ being held at Madras, Vellore and Lucknow. As part of this study, modification of the Disability Assessment Schedule (II) was carried out at the Madras centre. Certain items of the DAS were deleted and the rest were regrouped into 4 main areas of personal, social, occupational and global disability. This modified instrument called the Schedule for Assessment of Psychiatric Disability (SAPD) was administered to 30 patients each of the 3 groups of psychoses, neurotics and diabetics. It was found that the SAPD effectively discriminated the psychotic group from the other 2 groups. The authors recommend this instrument for measurement of disability in an outpatient psychiatric population.

SapF-mediated heme-iron utilization enhances persistence and coordinates biofilm architecture of Haemophilus

Non-typeable Haemophilus influenzae (NTHI) is a common commensal bacterium that resides in the human upper respiratory tract of healthy individuals. NTHI is also a known causative agent of multiple diseases including sinusitis, otitis media, as well as exacerbates disease severity of patients with cystic fibrosis and chronic obstructive pulmonary disease. We have previously shown that the Sap transporter mediates resistance to host antimicrobial peptides (AMPs) and import of the iron-containing compound heme. Here, we analyzed the contribution of the Sap structural ATPase protein, SapF, in these essential functions. In contrast to SapD, SapF was dispensable for NTHI survival when exposed to AMPs in vitro. SapF was responsible for heme utilization and recovery of depleted internal heme-iron stores. Further, a loss of SapF resulted in morphological plasticity and enhanced community development and biofilm architecture, suggesting the potential role of heme-iron availability in coordinating the complexity of NTHI biofilm architecture. SapF was required for colonization of the nasopharynx and acute infection of the middle ear, as SapF deficiency correlated with a statistically significant decrease in NTHI persistence in vivo. The Sleep and Pain Diathesis (SAPD) Model predicts that sleep quality is related to Fibromylagia (FM) outcomes such as disability and depression and that these relationships are mediated by both pain and impaired emotional dysregulation. The purpose of this paper is to provide a preliminary test of this model using cross-sectional data. 35 adult women, who had been living with FM for an average of 13 years, completed a battery of questionnaires that included reports of pain, sleep, affect, and disability. Consistent with this model, FM patients who reported more disrupted sleep also reported higher levels of psychological disability (i.e., BDI depression symptoms) and physical disability. Moreover, the trajectory of the relationship between sleep and pain appears to be mediated by cognitive processes such as increased pain helplessness and, thus, the relationship between sleep and disability appears to be mediated via pain. These data are consistent with the SAPD model, and lend support for the need to include sleep related factors as a critical contributor to our understanding of FM.

Behavioral manifestations of processing deficits associated with auditory processing disorder (APD) have been well documented. However, little is known about their anatomical underpinnings, especially cochlear processing. Cochlear delays, a proxy for cochlear tuning, measured using stimulus frequency otoacoustic emission (SFOAE) group delay, and the influence of the medial olivocochlear (MOC) system activation at the auditory periphery was studied in 23 children suspected with APD (sAPD) and 22 typically developing (TD) children. Results suggest that children suspected with APD have longer SFOAE group delays (possibly due to sharper cochlear tuning) and reduced MOC function compared to TD children. Other differences between the groups include correlation between MOC function and SFOAE delay in quiet in the TD group, and lack thereof in the sAPD group. MOC-mediated changes in SFOAE delay were in opposite directions between groups: increase in delay in TD vs. reduction in delay in the sAPD group. Longer SFOAE group delays in the sAPD group may lead to longer cochlear filter ringing, and potential increase in forward masking. These results indicate differences in cochlear and MOC function between sAPD and TD groups. Further studies are warranted to explore the possibility of cochlea as a potential site for processing deficits in APD.

Les kinases régulées par les signaux extracellulaires (ERK1/2) régulent une multitude de processus cellulaires, incluant la prolifération, la survie et la différenciation. Ces kinases représentent l’élément terminal de la voie ERK/MAPK, laquelle est activée dans près de 30% de tous les cancers humains et donc généralement perçue comme étant un effecteur critique de la progression tumorale. Cependant, une accumulation d’observations suggèrent que les kinases ERK pourraient également induire la suppression tumorale. Le but premier de cette thèse est de démontrer comment la signalisation par ERK peut contribuer à la suppression tumorale et de concilier les mécanismes impliqués avec son rôle dans la progression du cancer. Puisque nos travaux ont une incidence sur les bénéfices attendus de certaines thérapies actuellement en développement, le deuxième objectif de la thèse est de proposer de nouvelles stratégies thérapeutiques pour combattre le cancer. The innate immunity of vertebrates and invertebrates to microbial infection is mediated in part by small cationic peptides with antimicrobial activity. Successful pathogens have evolved mechanisms to withstand the antibiotic activity of these molecules. We have isolated a set of genes from Salmonella typhimurium which are required for virulence and resistance to the antimicrobial peptides melittin and protamine. Sequence analysis of a 5.7 kb segment from the wild-type plasmid conferring resistance to protamine contained five open reading frames: sapA, sapB, sapC, sapD and sapF, organized in an operon structure and transcribed as a 5.3 kb mRNA. SapD and SapF exhibited similarity with the 'ATP binding cassette' family of transporters including the bacterial Opp and SpoOK, involved in the uptake of oligopeptides; the yeast STE6, necessary for the export of a peptide pheromone; and the mammalian mdr, which mediates resistance to chemotherapeutic agents in cancer cells. SapA showed identity with other periplasmic solute binding proteins involved in peptide transport. The SapABCDF system constitutes a novel transporter for enteric bacteria and the first one harboring a periplasmic component with a role in virulence.

Descripció del recurs: el 5 de març de 2009; La síndrome d'aprimament post-deslletament (SAPD) és considerada com una malaltia porcina d'origen multifactorial en la qual el Circovirus porcí tipus 2 (CP2) és l'agent infecciós essencial. L'objectiu de la present tesi doctoral era expandir el coneixement en l'epidemiologia de la infecció per CP2 i el SAPD a través de la realització d'estudis de cas-control a nivell de camp. De manera general, es va investigar la potencial influència de la genètica del CP2, el moment d'infecció del CP2, els anticossos contra CP2 derivats de la immunitat maternal i la resposta humoral dels porcs contra el CP2, en la presentació de la SAPD. En el primer estudi (estudi I) es van estudiar seqüències de PCV2 de porcs amb diferent condició clínica i patològica. Els resultats van confirmar l'existència de dos genogrups principals i es va proposar la definició de dos genotipus de CP2 (1 i 2). La metodologia suggerida per definir els genotipus està basada en la distribució de la relació p-distància/freqüència de les seqüències de CP2 conjuntament amb anàlisis filogenètics de CP2. Die Grondwet van die Republiek van Suid-Afrika, 1996, vereis 'n fundamentele herassessering en transformasie van die wese en styl van polisiëring in Suid-Afrika. Dit moet vloei vanuit 'n stelsel waar die menseregte van die meerderheid Suid-Afrikaners tydens die apartheidsjare misken is, tot 'n stelsel wat die vertroue en respek van almal inboesem. Hierdie verandering vereis dat die fundamentele aannames van individuele polisiebeamptes ten opsigte van die organisasie en die omgewing waarin polisiëring funksioneer, sal transformeer. Op hierdie stadium, meer as elfjaar nadat die Suid-Afrikaanse demokrasie tot stand gebring is, is die sentrale vraag in polisiëring of die houdings van nuwelinge in die stelsel wel gemeenskapspolisiëring as die aanvaarde styl van polisiëring ondersteun.
